Ren Zhujianwei Granula exist good treatment of chronic atrophic gastritis (CAG).The subject is to establish CAG animal model to observe effects of Ren Zhujianwei Granula.Using rabbit serum of Ren Zhujianwei Granula to observe changes of apoptosis,P16,Fas and Survivin of gastric cancer cells.Part1,using N-Methyl-N-nitroso-N’-nitroguanidine(MNNG) and alcohol, then with irregular diet, broken down gas suffering and fatigue to establish CAG rats with Deficency of Spleen-Qi.To observe weight,temperature,stomach index and histopathological of CAG rats with Deficency of Spleen-Qi.To observe effects of Ren Zhujianwei Granula for CAG rats with Deficency of Spleen-Qi.Results:Weight of CAG rats with Deficency of Spleen-Qi was lower than normal significantly(p﹤0.01),and weight growth rate was lower than the normal significantly(p﹤0.01).Weight growth rate of Ren Zhujianwei Granula groups were higher than the model group significantly(p﹤0.01).Spleen weight of the model was lower than the normal significantly(p﹤0.01).Stomach index of the model was higher than the normal significantly(p <0.01).MNNG and alcohol may cause fluctuations in temperature of rats.Ren Zhujianwei Granula reduce fluctuations in temperature.Tip:Ren Zhujianwei Granula can increase weight of rats, stable temperature of rats.Part2,(1)Using CAG rats with Deficency of Spleen-Qi.To observe gastrin (GAS), somatostatin (SS) and.immunoglobulins(SIgA).Results:GAS and SS of CAG rats with Deficency of Spleen-Qi was lower than normal significantly(p﹤0.05).After treatment,GAS and SS of Ren Zhujianwei Granula groups were higher than CAG rats with Deficency of Spleen-Qi significantly(p﹤0.05).SIgA of CAG rats with Deficency of Spleen-Qi was lower than normal significantly(p﹤0.05).SIgA of Ren Zhujianwei Granula groups were higher than CAG rats with Deficency of Spleen-Qi significantly(p﹤0.05).Tip:Ren Zhujianwei Granula can adjust abnormal changes in GAS, SS and SIgA to normal levels.(2)Using CAG rats with Deficency of Spleen-Qi.To observe endothelin(ET), nitric oxide synthase(NOS) and.motilin (MTL).Results:ET and NOS of CAG rats with Deficency of Spleen-Qi was higher than normal significantly(p﹤0.05).After treatment,ET and NOS of Ren Zhujianwei Granula groups were lower than CAG rats with Deficency of Spleen-Qi significantly(p﹤0.05).MTL of CAG rats with Deficency of Spleen-Qi was higher than normal significantly(p﹤0.01).MTL of Ren Zhujianwei Granula groups were lower than CAG rats with Deficency of Spleen-Qi significantly(p﹤0.05).Tip:Ren Zhujianwei Granula can adjust abnormal changes in ET,NOS and MTL to normal levels. (3)Using CAG rats with Deficency of Spleen-Qi.To observe matrix metalloproteinase2(MMP-2),epidermal growth factor receptor(EGFR), cyclooxygenase2(COX-2).Explore the treatment of Ren Zhujianwei Granula for CAQas well as the mechanism of that prevention of gastric cancer.Results:MMP-2,EGFR and COX-2of CAG rats with Deficency of Spleen-Qi was higher than normal significantly(p<0.05).After treatment,MMP-2,EGFR and COX-2of Ren Zhujianwei Granula groups were lower than CAG rats with Deficency of Spleen-Qi significantly(p<0.05).Tip:Ren Zhujianwei Granula can adjust abnormal changes in MMP-2,EGFR and COX-2to lower levels.Part3,exploreing the effect of Ren Zhujianwei Granula serum on malignant of GES-1transformed cells induced by MNNG(MC).To observe proliferation,P16Fas,Survivin and apoptosis of MC.Results:Proliferation of Ren Zhujianwei Granula serum groups were lower than NS group significantly(p<0.05).P16of Ren Zhujianwei Granula serum groups were higher than NS group significantly(p<0.05).Ren Zhujianwei Granula serum may inhibit proliferation of MC.Apoptosis of Ren Zhujianwei Granula serum groups were higher than NS group significantly(p<0.05).Fas of Ren Zhujianwei Granula serum groups were higher than NS group significantly(p<0.05).Survivin of Ren Zhujianwei Granula serum groups were lower than NS group significantly(p<0.05).Tip:Ren Zhujianwei Granula serum may improve the expression of P16of MC. Ren Zhujianwei Granula serum may improve the expression of Fas and reduce the expression of Survivin causing apoptosis of MC.Conclusion:Using chemical carcinogen and integrated approach may establish CAG rats with Deficency of Spleen-Qi model successfully.Model were addicted to lying, curled get together,hair haggard,struggling to crawl weakness and so on.Lesions occur in pathological of stomach tissue, gastrointestinal hormones,NOS,ET,SIgA,COX-2,MMP-2,EGFR et al.Ren Zhujianwei Granula may adjust pathological of stomach tissue,temperature, gastrointestinal hormones,NOS,ET,SIgA,COX-2,MMP-2,EGFR et al.Rabbit serum of Ren Zhujianwei Granula may inhibit proliferation of cancer cell,and promote apoptosis of them.
