Objective : to investigate the expressive changes of TLR4, IKK-βin asthma rats with spleen deficency, whose influence on air passage inflammation, and discusse the effect of"TCM prescription for reinfocing Pi Qi"on regulating the TLR4, IKK-βand IL-8 in asthmatic rats'lung tissues to further reveal the molecule mechanism of asthmatic attack and the therapeutic effec of Bupiyiqi prescription.Method : SPF male wistar rats were randomly divided into normal control group(A),asthmatic group(B), asthmatic with spleen deficiency group(C), asthmatic group by TCM treated(D), asthmatic with spleen deficiency group by TCM treated(E), asthmatic group by western medicine(F), asthmatic with spleen deficiency group by western medicine treated(G). Rats were established into spleen deficency model via appling the methods of over-fatigued, bitter cold herb feed, and unregular diet, for 10 days. Rats were established into asthmatic model by sensitized and provoked ovalbumin. At the 11th day, model rats were stated to be treated by Bupiyiqi medicine, or western medicine-dexamethasone, for 21 days. The protein expression level of mRNA of TLR4 and IKKβin lung tissue were determined by Polymerase Chain Reaction (RT-PCR); the result of interleukin eight (IL-8) in bronchoalveolar lavage fluid (BALF) were detected by enzymes linked immunosorbent assay(ELISA).Result :①the expression of mRNA of IKKβin lung tissues: B and C was significantly higher than A(P<0.01); C was significantly lower than B group (P<0.05); E and G was significantly lower than C(P<0.01)without difference between those two groups; so as the D and F.②the expression of protein of IKKβin lung tissue : C was significantly higher than A group(P<0.01); C group was significantly lower than B group (P<0.01). C was highter than B (P<0.05); E and G was significantly lower than C(P<0.01) and E was lower than G(P<0.05); so as the D and F③the level of IL-8 in BALF: B and C was significantly higher than A(P<0.01), and C was higher than B(P<0.01); C group was significantly lower than B group(P<0.01); E and G was significantly lower than C(P<0.01) without differences between those two groups; so as the D and F.④the expression of mRNA of TLR4 in lung tissues: B and C was significantly higher than A(P<0.01); C was significantly lower than B group (P<0.01); E and G was significantly lower than C(P<0.01)without difference between those two groups; so as the D and F.⑤the leve of IL-8 in BALF: the level of IL-8 in BALF: B and C was significantly higher than A(P<0.01), and C was higher than B(P<0.01); C group was significantly lower than B group(P<0.01); E and G was significantly lower than C(P<0.01) without differences between those two groups; so as the D and F but with differences between groups.Conclusion:①spleen deficency can increase the mRNA expression of IKKβand TLR4 signicantly in lung tissue of asthmatic rats,so as the level of IL-8 in BALF.②the expression of IKKβand TLR4 is positive correlation, and have the same relationship with IL-8. Bupiyiqi prescription and dexamthason can reduce the the mRNA expression of IKKβand TLR4, consequently decrease the leve of IL-8 via③"TCM prescription for reinfocing Pi Qi"and dexamethasone could inhibit expression of IKKβand TLR4 efficiently and reduce the level of IL-8.
