Objects:To investigate the changes of cardiac function in septic rats and the effects from Xuebijing injection.Method:Cecal ligation puncture (CLP) was performed to induce sepsis rats. Male Wistar rats were adaptive fed for one week,fasted for12hours before operation.After anesthesia,the abdominal incision was made about2cm. The cecum was exposed and its roots was ligated.The cecum was penetrated twice to formate intestinal fistula.Two drainage strips were placed to prevent pinhole closure. The cecum is placed back to the abdominal cavity.Then the abdominal incision was sutured. Animals were treated with normal saline (10mL) to prevent shock by subcutaneous injection. In sham operation,only open-close abdomen was performed. Seventy-eight rats were selected and randomly divided into four groups.①control group(n=6),animals were observed by echocardiography;②sham operation group(n=24), animals were treated with normal saline (4mL/kg each) by intravenous injection at2,12,24,36,48and60hours and observed by echocardiography at6,24,48,72hours after operation;③CLP group (n=24), animals were treated with normal saline (4mL/kg each) by intravenous injection at2,12,24,36,48and60hours,and observed by echocardiography at6,24,48,72hours after operation;④XBJ group (n=24), animals were treated with Xuebijing injection (4mL/kg each) by intravenous injection at2,12,24,36,48and60hours,and observed by echocardiography at6,24,48,72hours after operation.The blood samples of each group were collected at various time points from abdominaly aorta. Serum levels of cardiac troponin T (TnT) was detected by euzymelinked immunosorbent assay (ELISA).Sample of heart was harvested aseptically. The histopathologic changes of myocardial tissue were observed and the myocardial histopathologic score was counted by optical microscope.Result:1.The changes of cardiac function in septic rats and the effects from Xuebijing injection In CLP group LVEDD was increased at6,24,48h (P<0.05), restored to normal at72h (P>0.05). Compared with CLP group,no significantly differences of LVEDD were found in XBJ group (P>0.05). Compared with the normal group and sham group, HR was increased from6to72h after CLP (P<0.05),and had no effect after using Xuebijing injection (P>0.05). Moving amplitude of left ventricular posterior wall tended to decrease in CLP group(P>0.05). Compared with the normal group and sham group, moving amplitude of interventricular septal were decreased markly after CLP at48h (P<0.05), and had no effect after using Xuebijing injection (P>0.05). LVEF was significantly decreased from6to72h in CLP group (P<0.05), and it was markedly increased in XBJ group (P<0.05). In CLP group Ea/Aa was decreased at6,24,48h (P<0.05), restored to normal at72h (P>0.05). Compared with the CLP group, Ea/Aa was markedly increased in XBJ group at6,24,48h (P<0.05).2. The histopathologic changes of myocardial tissue in septic rats and the effects from Xuebijing injection Compared with the normal group and sham group, the myocardial histopathologic score was increased from6to72h after CLP (P<0.05), and it was markedly decreased in XBJ group (P<0.05).3. The Changes of the serum concentration of TnT in septic rats and the effects from Xuebijing injection Compared with the normal group and sham group, the serum concentration of TnT was increased from6to72h after CLP (P<0.05), and it was markedly decreased in XBJ group (P<0.05).4. The correlation between myocardial function and myocardial histopathologic score, TnT and LVEDD In CLP and XBJ group,there exists negative correlation respectively between LVEF and myocardial histopathologic score, TnT and LVEDD with statistical significance (P<0.05).Conclusion:1.There are structural changes and dysfunction of heart in septic rats, showing increased left ventricular end-diastolic dimension, interventricular septal and left ventricular posterior wall motion disorders, left ventricular systolic and diastolic dysfunction.2.There are myocardial injury in septic rats, showing increased serum level of TnT and myocardial pathologic score. And Injury of myocardial cells contribute to the systolic and diastolic dysfunction.3. It is illumineted that Xuebijing injection could protect cardiac function in septic rats,through reducing the myocardial injury.
