Object:To observe the levels of serum I-FABP,MLT,GAS of the acute liver failure rats and to analysis the relationship between these indicators and gastrointestinal dysfunction of these rats,and providing theoretical and experimental basis for the early prediction of gastrointestinal dysfunction with acute liver failure.Method:85female healthy clean rats were randomly divided into ALF model group (n=50) and normal control group (n=35), all the rats were respectively adaptive fed for three days.ALF model rat were injected with350mg/Kg TAA (paired40g/L with NS) by intraperitoneal,injected again after24h,and ALF model of rats were completed.The normal control group were injected with the same amount saline by intraperitoneal at the same time.The rats’blood were taken at24h,36h,48h after completed injection,and detected the liver function including ALT,AST,TBIL,and detected the levels of I-FABP,MLT,GAS by enzyme-linked immunosorbent assay. The right lobe liver,the gastric antral and2cm proximal duodenum were collecated at the same time.All the samples were fixed by the buffer solution of40g/L formaldehyde,paraffin-embedded,sliced,stained by HE,and observed the pathological changes of the liver,gastric and duodenum tissue under the light microscope. SPSS16.0statistical software were used for statistically analyzing the experimental data.The measurement data were shown with thex±s, the difference of indexes between different group were test with the single-factor analysis of variance, the correlation of two index were test with Pearson correlation analysis(two-sided test), P <0.05was significant difference.Result:1. The clinical characterytics of two groups of rats:The control group:rats were activating freely,responsive,eating freely, defecated and urinating normally, and all of them were living animals.The ALF model group:rats were eating reduced,even refused to eat,were listlessness,drowsiness,coma,and unresponsive,the activity of them were reducing, their abdominal were bulging,seen obvious gastrointestinal-type et al,such performance as hepatic encephalopathy and gastrointestinal dysfunction. 2. The comparison of liver function between the ALF model group and control group.Compared with the control group, the level of ALT,AST and TBIL of the ALF model group rats were significantly increased each time point at24h,36h,48h (ALT(U/L)24h:346.81±127.04/50.15±11.94,36h:182.07±72.52/50.24±11.51,48h:98.36±31.84/49.82±9.78; AST(U/L)24h:1311.25±552.20/161.26±36±.49,36h:744.64±290.00/164.33±39.91,48h:440.66±123.93/165.20±42.81;TBIL(mmol/L)24h:6.54±2.32/3.79±1.15,36h:8.45/3.38/3.75±1.12,48h:13.20±4.45/3.76±1.22].(P<0.01),and the level of the ALT,AST were gradually decreased,and the level of TBIL were increasingly with the time,bile enzyme separation were detected.3.The comparison of the level of I-FABP,MLT and GAS between the ALF model group and control group.Compared with the control group,the level of I-FABP(ng/L) of the ALF model group rats was significantly increased each time point at24h,36h,48h (24h:14.41±5.07/10.48±2.29,36h:18.60±5.57/10.46±4.14,48h:22.63±6.86/9.60±3.67)(P<0.05).And the level at48h was higher than that at24h (P<0.01).The level of MLT(ng/L) was significantly reduced at each time point(24h:135.50±30.15/265.10±44.35,36h:125.84±42.00/253.19±49.18,48h:144.19±40.74/262.70±58.48)(P<0.05). The level of GAS(ng/L) was significantly increased at each time point(24h:2.15±0.88/1.45±0.48,36h:2.37±0.89/1.52±0.48,48h:3.45±1.57/1.58±0.83)(P<0.05).And the level at48h was higher than that at24h (P<0.01).4.The pathological changes of the liver,gastric antrum and duodenum of the two groups:There were no abnormalities found in the liver,gastric antrum and duodenum tissue of the control group rats observed by light microscopy.The ALF model group:the tissue structure of rats liver were unclear,hepatic lobule were disordered,the liver cell were high swilling and ballooning degeneration, Inflammatory cells infiltrated around the portal area,and the necrosis of liver cell around the central venous were most obvious(focal necrosis and large areas of necrosis).Epithelial cells and Intestinal villi of the stomach and duodenum mortified and shed.Conclusion: 1.In this experiment, rat model of acute liver failure with gastrointestinal dysfunction by injecting TAA (paired40g/L with NS)350mg/Kg by intraperitoneal for2times interval24h were successfully set up.2.The level of serum I-FABP of the ALF model group rats was significantly increased,suggesting that I-FABP can be used as monitoring indicators of acute liver failure with gastrointestinal mucosal ischemia and gastrointestinal dysfunction.3.The level of serum MLT was significantly reduced,and the level of serum GAS significantly increased,MLT and GAS might play a role in the mechanism of acute liver failure with gastrointestinal dysfunction,and the levels of these two protein could be used as one of the indicators of gastrointestinal dysfunction.
