Regulation Of Extraction Of Semen Coicis On The Expression Of MMP1, MMP3Induced By Ultraviolet B Radiation In The Human Skin Fibroblast

Objective:To investigated the expression of matrix metallo proteinase1,3(MMP1, MMP3) of human skin fibroblasts induced by UVB radiation, and to explore the protective effect of KLT on the expression of matrix metallo proteinase1,3(MMP1, MMP3) of human skin fibroblasts induced by UVB radiation.Methods:Hunman skin fibroblasts were cultured and grouped. MTT were performed to detected the effect of KLT on the proliferation of fibroblasts. Then, assessed the effects of KLT treatment on mRNA and protein expressions of MMP1, MMP3in UVB radiated fibroblasts.Results:Fibroblasts cells could proliferate in a short time.3-5days is logarithmic growth period of fibroblasts and5-7days is a platform period. After the sixth generation, the cells gradually lose energy and shown an irregular shape. KLT exhibited no inhibitory effect on the proliferation of fibroblasts. The expression of MMP1, MMP3mRNA and protein in different dose UVB radiated fibroblasts expressed significant difference (p<0.05), especially in30mJ/cm2UVB group. KLT treatment exhibited down-regulation on the expression of MMP1, MMP3mRNA and protein in30mJ/cm2UVB radiated fibroblasts (p<0.05).Conclus i ons:Hunmen skin fibroblasts could be gotten in a short time, and the second generation to the fifth generation have great proliferation capacity and they can grow steadily, so they are appropriate to be used in the experiment. KLT have no influence on the proliferation of fibroblasts and can effectively down-regulate the expression of MMP1, MMP3mRNA and protein of the fibroblasts. So, we speculate KLT could remit photo damaging induced by UVB.
