[Objective]All of the AD model mice were induced by 2,4-Dinitrochlorobenzene?DNCB?and olive oil.After modeling,To investigate the therapeutic effect of Semen Coicis Extract?SCE?and the matrix of SCE on atopic dermatitis in mice,as well as,the effects of skin lesions,serum IFNlammatory cytokines,aquaporins 3 and toll-like receptor 2 and 4 expression were studied.Explore its possible mechanism of action.[MethoDs]Forty female SPF BABL/c mice were divided into five groups randomly and equally:AD model without intervention group 8,the SCE intervention group,the matrix of SCE-control group 8,AD model group 8,blank group 8.On the day before the program,All of the mouse's back was applied with a 10%sodium sulphide solution?about 5 x 5mm2?.In the three days prior to the experiment,1%2,4-dinitrochlorobenzene?DNCB?and acetone and olive oil were used to stimulate the back and ear of other than the blank group.After that,0.1%2,4-dinitrochlorobenzene?DNCB?and acetone and olive oil were used to stimulate the back and ear of other than the blank group for 12 times.After modeling,the AD model group and the black group were sacrificed by breaking the neck after intraperitoneal injection of chloral hydrate.AD model without intervention group were not treated,so that they could be repaired naturally.The two treatment groups were treated each day for twenty-eight days.The skin lesion were detected before and after treatment.After the experiment,the mice were anesthetized,and the mice were executed by the method of broken neck after intraperitoneal injection of chloral hydrate.The thickness gauge was used to detect the thickness of left ear.The IFNiltration of dermatitis cells was observed by pathological section.The expression changes of AQP3 and Toll-like receptor two and four were detected by immunohistochemistry.The levels of IgE,interlukin-4 and interferon-r were measured by ELISA.[Results]The skin lesions of the mice in SCE intervention group was basically subtracted from erythema,exudation and hyperplasia.The skin lesions in AD model without intervention group and the matrix of SCE-control group showed slight erythema,dry and partial scabs.In the SCE intervention group,the thickness of the ear was less than that of the AD model without intervention group?P<0.05?.The pathological results of blank group showed normal epidermis,dermis structure and no IFN lammatory cell IFNiltration.In AD model without intervention group and the matrix of SCE-control group,mild hyperplasia of epidermis,intercellular edema,dermal neutrophils and lymphocyte IFNiltration were observed.The epidermis of the SCE intervention group was slightly thicker than group blank group,with a small amount of IFNlammatory cells IFNiltrating in the dermis.In SCE-control group,the expression of AQP3 spine was lower than that of group AD model without intervention group.Immunohistochemical findings indicated that the expression and localization of TLR2,TLR4 and AQP3 were changed.Compared to the AD model without intervention group,The levels of IgE and il-4 in the treatment group were decreased,and the IFN-r level was higher?P<0.05?.[Conclusion]Our study suggested that topically SCE is an effective medication in the treatment of AD in mice.It is possible that the expression of serum IgE,IL-4 and IFN-levels and the expression of AQP3,TLR2 and TLR4 may be affected.
