| Objective : By observing the influence of different frequencies' electric acupuncture and coicis semen to rats after hemi-section Spinal Cord Injury on morphology and ethology, the experiment intend to discover the functional mechanism of them, to screen better treating parameter of the electric acupuncture and to find out the repairing function of coicis semen to injured spinal. It provides the experimental basis for clinical practice.Method: Select the Wistar rats for this experiment. Construct the hemi-section Spinal Cord Injury model. And divide rats into four groups randomly. They are the model group, low-frequency (5Hz) electric acupuncture group, high-frequency (lOOHz) electric acupuncture group and coicis semen group. In addition, the experiment takes the normal group and sham operation group as control group. Treatments are carried on in 3 hours after the operation. Zhiyang, Xuanshu, Huantiao, Zusanli of the suffering hind iimb are acupunctured in the electric acupuncture groups. They are carried on every other day and 1 Omin every time for 8 weeks. The treatment to the coicis semen group is the intraperitoneal injection of the water extraction of coicis semen. Every rat is injected 0.4ml every other day. The evaluation of the ethology goes on in 11 time points. They are everyday of the first week after operation, and in the second, 4th, 6th and 8th week. The evaluation scoring is based on the BBB (the Basso—Beattle—Bresnahan) evaluation method. The rats are executed after 8 weeks. The detection of Motion Evoked Potential is done before making model, instantly after operating and before executing. Observe HE (hematoxylin and eosin) stain, GFAP (anti- glial fibrillary acidic protein) immune and histochemical stain. Survey the ultrastructure under electron microscope.Results:1. In 11 time points, the scores based on BBB of the therapeutic groups are all significantly higher than that of the model group (P<0.01, P<0.05). However, indifferent time points, it shows different therapeutic results of different frequency groups. The score of the coicis semen group is the highest in one day after operation, the score of the low-frequency (5Hz) electric acupuncture group is the highest during early stage (3-7d), and the score of the high-frequency (lOOHz) electric acupuncture group is the highest during advanced stage (4-8W). And there is no statistics significance among these groups.2. The wave form of the spinal cord, which is hemi-transected, disappears instantly after operating. In 8th week after operation, comparing with model group, the wave amplitude changes of the lower frequency group are significantly higher (P<0.01). However, the latent time has no statistics significance (P>0.05). In higher frequency group and the coicis semen group, the wave amplitude changes and latent time all have statistics significance (PO.01). The comparing among the electric acupuncture groups on the latent time and wave amplitude changes shows that the effect of high-frequency (lOOHz) electric acupuncture group is significantly better than that of low-frequency (5Hz) electric acupuncture group (P<0.01,P<0.05) .3. In 8th week after operation, the positive GFAP expressing area in spinal cord of the normal group is the smallest. And that of the sham operation group has no significant difference comparing with normal group (P>0.05). The biggest expressing one is model group. Comparing with normal group and sham operation group, it has significant difference (P<0.01). And the positive GFAP expressing area of the therapeutic groups are both significantly bigger than that of the model group and the normal group (PO.01) .The results of electron microscope show that the ultrastructure of high-frequency (lOOHz) electric acupuncture group is the best. The coicis semen group take&the second place. The low-frequency (5Hz) electric acupuncture group takes the third place. And the model group is the worst.Conclusion: The electric acupuncture therapy and coicis semen can all effectively promote the recovery after Spinal Cord Injury. The curative effect of the low-frequency (5Hz) electric acupuncture group is the best during early stage, and the effect of the high-frequency (lOOHz) electric acupuncture group is the best during advanced stage. |
