Objective:To detect the expression of mammary serpin (Maspin),basic fibroblast growth factor (bFGF) in patients with chronic cervicitis, cervical intraepithelial neoplasia and cervical cancer and explore the relationship between Maspin,bFGF and high-risk human papilloma virus (high-risk type HPV) infection.Methods:Cervical paraffin tissue samples were collected (n=166) in which106cases were in high-risk type HPV positive group (chronic cervicitis20cases,CINⅠ grade20cases,CINⅡ grade20cases,CINⅢ grade20cases,cervical cancer26cases);60cases were in HPV negative (HPV-) group(CINⅠ grade35cases,CINⅡ grade20cases,CINⅢ grade5cases). Expression of maspin,bFGF protein was detected by Immunohistochemical SP method.Results:1.In High-risk type HPV (+) chronic cervicitis, CINⅠ, CINⅡ, CINⅢ and cervical cancer groups:①The differences between the positive rate of Maspin protein expression in the five groups were statistically significant and they were gradually decreased (x2=37.78, P<0.001); The rate of Maspin protein positive expression in cervical cancer was statistically lower than the other groups (P<0.05or P<0.01); The rate of Maspin protein positive expression in CINⅠ,CINⅡ, chronic cervicitis was statistically higher than that in CINⅢ group (P<0.05). Maspin protein expression was negatively correlated with the severity of lesion (rs=-0.573,P<0.001).②The differences between the positive rate of bFGF protein expression in the five groups were statistically significant and they were gradually increased (x2=18.94. P=0.001); The rate of bFGF protein positive expression in cervical cancer was statistically higher than those in CINⅠ,CINⅡ, chronic cervicitis groups (P<0.05); The differences between the positive rate of bFGF protein expression in the there CIN groups were not statistically significant(P<0.05); The rate of bFGF protein positive expression in CINⅢ group was statistically higher than that in chronic cervicitis group (P<0.05). bFGF protein expression was positively correlated with the severity of lesion (rs=0.416, P<0.001)2. In HPV(-)CINⅠ, CINⅡ, CINⅢ groups:The positive rates of Maspin protein were gradually lower among CIN groups but the differences between groups were not statistically significant (χ2=1.278,P=0.528);The positive rate of bFGF in CIN groups were gradually increased but the differences were not statistically significant (χ2=0.163, P=0.922).3. In the five High-risk type HPV (+) groups,the relationship of Maspin protein expression and bFGF protein expression was negatively correlated (r=-0.325, P<0.001).In the High-risk type HPV (+) and HPV(-) CIN groups,The relationship of Maspin protein expression and high-risk type HPV infection was negatively correlated (r=-0.205,P=0.024) and The relationship of bFGF protein expression and high-risk type HPV infection was positively correlated (r=0.189, P=0.038), by using Spearman correlation analysis.Conclusion:Maspin may play a certain suppressive role in the development of cervical cancer but bFGF may play a promoting role in the process; High-risk type HPV may degrade Maspin protein expression and upgrade bFGF protein expression through some way, and result in the cervical leision from chronic cervicitis to CIN to cervical cancer.
