Objective:(1) To sudy the expression of Maspin,Caspase-3 and COX-2 in cervical carcinoma.To evaluate the correlation between them and clinico-pathologic variables in these cervical carcinoma patients. (2) To study the effect of sinomenine and DDP on the expression of Maspin,Caspase-3,COX-2 protein in human cervical cancer cell line Hela.(3) To investigate the effect of sinomenine and DDP on the proliferation and apoptosis of human cervical cancer cell line Hela .Methods:(1) The expression of Maspin,Caspase-3 and COX-2 in cervical carcinoma were examined by immunohistochemistry method. (2) The effect of sinomenine and DDP on the expression of Maspin,Caspase-3,COX-2 protein in human cervical cancer cell line Hela were analyzed by immunocytochemistry.(3) The effect of sinomenine and DDP on the proliferation and apoptosis of human cervical cancer cell line Hela were analyzed by cell cultured method,MTT assay and flow cytometry(FCM). Results:(1).By MTT assay, sinomenine,DDP and both inhibited proliferation in a dose- and time-dependent manner(P<0.05),accelerated the apoptosis of Hela in a dose-dependent manner(P<0.05).(2)By immunocytochemistry ,when incubated with sinomenine,DDP and both, it was found that the negative expression of Maspin,Caspase-3 and were increased and the positive expression of COX-2 were statistically declined in vitro.(3) By immunohistochemistry method, reductionor absence of Maspin expression was found in 58%(29/50)of the tumors. Positive staining of Caspase-3 was detected in 43 of 50(86%) cervix carcinoma, Caspase-3 immuno activity was observed in 21of 50 (42%) cervix carcinomas.The expression of Maspin was irrelevant with histological grade and lymlph nodes metastasis.The expression of COX-2 correlated significantly with histological grade and clinical stages(P<0.05).Loss of caspase-3 increased with histological grade.There was a positive correlation between Maspin and Caspase-3(P<0.05),and a negative correlation between Maspin and COX-2 (P<005).Conclusion:(1) Both sinomenine and DDP inhibited the proliferation and induced the apoptosis of Hela cell lines in vitro.(2) By immunocytochemistry,when incubated with sinomenine and DDP,it was found that the expression rates of Maspin,Caspase-3 were increased and the positive expression rates of COX-2 were statistically declined.(3)The unexpression of Maspin,Caspase-3 and the overexpression of COX-2 may play important roles in carcinogenesis and development of cervical carcinoma,they were valuable to further understand the biolodigal behavior of cervical carcinoma.
