| The plant Acanthopanax gracilistylus W. W. Smith belongs to Araliaceae which is widely distributed in China, and its dried cortex are listed officially in the Chinese pharmacopoeia as Cortex Acanthopanacis(named as Wujiapi), Which has been used as medicine for the treatment of paralysis, arthritis, rheumatism, lameness and liver disease. Besides its roots, leaves and fruits are also used as medicines. Though Acanthopanax gracilistylus is widely distributed in China, wild resouces reduced year after year because of the ecnomic development. Even worse, it is a great confusion for clinic use and market sales, for example, Xiangjiapi is used as a substitute for Acanthopanax gracilistylus. So, we studied the ecology distributing, reproduce characteristic, chemical consitutents and evaluted the quality of different populations of Acanthopanax gracilistylus from Jiangsu Province. Results obtained were as follows:1. Determination of chlorophyll content, photosynthesis, leaf morphology and microstructure observation of different materials in Acanthopanax gracilistylus of Tieshansi, Baohuashan, Tangshan population was done in this research. Different populations of Acanthopanax gracilistylus were significantly different in leaf morphology; Tangshan population had the highest content of chlorophyll, leaf from3populations had shown typical characteristics of shade plants. The diurnal variation of net photosynthesis rate in these three populations was a single peak cure. The light compensation point and light saturation point amounted to24μmol·m-2·s-1and555μmol·m-2·s-1in Tangshan population, while21μmol·m-2·s-1and348μmol·m-2·s-1in Tieshansi,18μmol·m-2·s-1and309μmol·m-2·s-1in Baohuashan. Among the three populations, the Tangshan population had the highest Pn and CE. 2. For the purpose of developing and utilizing Acanthopanax gracilistylus, we had made chemical studies on the constituents of the cortex of Acanthopanax gracilistylus. The chemical components were isolated and purified by silica gel column, ODS C-18chromatography, MDS chromatography.7components were isolated, and the structures were elucidated on the basis of physic-chemical properties and spectral data. These7components are stearic acid,1-sesamin, β-sitosterol, β-sitosterolglucoside,[(-)-kaur-16-en-19-oic acid], syringin.3. Using Acanthopanax gracilistylus as the materials, orthogonal design was used to get the best method, anthrone-sulfuric acid spectrophotometric to determine total polysaccharide content. The optimal extraction conditions were:temperature of80℃, time of2.5h, and the ratio of material to solvent was1:20. The content of polysaccharides in cortex, bark and leaf respectively reaches15.66%,8.94%and16.54%. Polysaccharide content in different parts and confusing products were significantly different.4.12kinds of mineral elements were ananyzed in Acanthopanax gracilistylus. The concentration of Ca and K were about18502.1μg·g-1and13979.3μg·g-1which were seperately higher than the content of P and Na. Content of mineral elements were different from different populations and parts.5. Using HPLC method for different populations of Acanthopanax cortex and different parts to determine the syringin content. The syringin content of Acanthopanax cortex were significantly different from different populations, Tieshansi population as the highest which was0.852mg·g-1. The content of the syringin from high to low was as follows:cortex> bark>leaf>fruit. |
