Objective Cerebral vascular disease morbidity, mortality and disability rates are very high, as the three leading cause of death among all disease. Die each year from stroke patients of about1500000people, of which75%?~85%for ischemic cerebral vascular disease. Focal ischemic cerebral vascular disease by vascular congestion caused by local cerebral blood flow decreased, resulting in tissue hypoxia and cell death, formed by the central necrosis and peripheral areas of the ischemic penumbra consisting of local infarction lesion, in which the cell necrosis and apoptos is coexist, apoptosis occurs mainly in the ischemic penumbra. At this time the body to make a response to maintain oxygen balance and steady state to resist the adverse factors, one of the important mechanisms that induce neovascularization, improved collateral circulation increases blood flow to the brain oxygen metabolism to compensate enough to save the dying neural cells. Research shows, cerebral vascular occlusion after ischemic vascular hypertrophy, hyperplasia of the extent and rang is directly related to the penumbra blood flow improvement, affect neuronal physiological function recovery. Cerebral vascular occlusion, ischemic vascular proliferation, spontaneous formation of coronary collateral circulation trend, but not enough to compensate the original blood supply, ischemia not corrected. Mentor previous studies showed that electro-acupuncture on promotion of focal cerebral ischemia in the rat brain angiogenesis has certain effect, but its effect on angiogenesis factor. The topics to MCAO and reperfusion in rats as a model, angiogenesis and apoptosis from the point of view of electro-acupuncture to effectively protect and improve the role of ischemic brain damage in target and its mechanism.Methods Using Longa arteries modified suture method in making rat MCAO model, selection of120healthy adult SD rats were randomly divided into normal control group, sham-operation group, MCAO model control group (lhour ischemia and reperfusion1/2d,1d,2d,4d,7d five sub-groups), electro-acupuncture group (lhour ischemia and reperfusion1/2d, Id,2d,4d,7d five sub-groups), the former two groups each group10only the latter two groups for each sub-group of10rats. The normal group, sham operation group, model group without treatment, electro-acupuncture group of electro acupuncture "Baihui"、"Shuigou"、"Zusanli", every other12hours treatment time. By reperfusion at different time points for subjects taken before and after the2mm flat part of the optic chiasm, using immunohistochemical S-ABC method, optical microscope to observe the rat ischemic brain tissue microvascular density and HGF, HIF-1α expression level; all the data are entered into the computer in order to SPSS18.0software statistical package processing, experimental results are the mean standard deviation indicated,Groups were analyzed using t-test. Result (1) Behavioral score observed:model group and acupuncture group showed obvious symptoms of nerved damage, electro-acupuncture group nerve symptom score after reperfusion1d,2d,4d,7d group and model group, significantly reduce the symptoms.(2) HE staining:normal group and sham operation group did not see neuronal degeneration or necrosis and glial cell hyperplasia. Model group of nerve cell necrosis area in general than the electro-acupuncture group, but glial cell proliferation than electric acupuncture; electro-acupuncture group hyperplasia faster and model group.(3) Microvessel density:normal group and sham operation group of brain tissue in rats with almost no positive expression of CD31, CD31model group of microvessels in ischemia reperfusion Id rose rapidly, following the rise7d reperfusion. Ischemic zone around the positive expression of CD31increased significantly, around the infracted area of small vascular endothelial cell cytoplasm was brown, densely arranged, showed a strong positive expression. Electro-acupuncture group and model group expression of basically the same, but the more widely, in the corresponding period of time compared with model group, electro-acupuncture group had significant difference (p<0.05orP<0.01)(4) HGF in rat cerebral ischemia cortex expression:normal group and sham operation group of brain tissue in rats with only a small amount of HGF positive expression increased significantly, compared with normal group and sham operation group, the difference has statistical significance (p<0.05), which peaked at7d. In the corresponding period of time, the electro-acupuncture group positive expression was higher than in the model group, the difference was statistically significant (p<0.05orP<0.01), description of electro-acupuncture can increase cerebral ischemia reperfusion rat brain HGF expression.(5) HIF-1a in rat cerebral ischemia cortex expression:normal group and sham operation group of brain tissue in rats with only a small amount of positive expression of HIF-1a model group, electro-acupuncture group of the positive expression increased significantly (p<0.05), which peaked at7d; electro-acupuncture group positive expression was higher than that in the model group, the difference was statistically significant (p<0.05orP<0.01), description of electro-acupuncture can up regulate brain in rats with cerebral ischemia and reperfusion on the expression of HIF——1a.Conclusion Electro-acupuncture "Baihui"、"Shuigou"、"Zusanli" points able to focal cerebral ischemia—reperfusion injury in rat brain has a protective effect-can effectively improve after rat cerebral ischemia neurological symptom scores, can promote the ischemic area and the surrounding brain angiogenesis. The role of the mechanism may provide electro-acupuncture increased ischemic areas of rats with HGF, HIF-1αup regulates the expression of, thereby facilitating the ischemic area and around the cerebral vascular proliferation.
