ObjectUsing MTT,flow cytometry and Western-blot analysis to detect of compound angelica injection on proliferation and apoptosis,and expression of bcl-2and bax protein in myofibroblast(MFB),exploring the mechanisms of these agents.Searching for a effective drug to treat oral submucous fibrosis.MethodFB and MFB were treated with compound angelica injection detected by MTT colorimetric assay.Cell apoptosis percentage and the expression level of bcl-2and bax protein after different time and concentration course of compound angelica injection were determined by flow cytometsy and western-blot analysis reapectively.Result1. Compound angelica injection could inhibit the proliferation of FB and MFB in a dose and time dependent manner.,inhibit rate of MFB is higher than FB. 2. Compound angelica injection administered individually for24h,48h,72h,the apoptotic rate of MFB slightly increased after different time and concentration course.Expet there was no statistics significance between5mg/ml dose in24h group and control group.there were statistics significance (P<0.01) for other groups/these agents markedly improved apoptosis rate in a manner depending on time and concentration.3. Compound angelica injection administered individually for24h,48h,72h,the expression bax was upregulated,and the expression of bcl-2was downregulated. Compound angelica injection induces apoptosis in myofibroblasts by down-regulating the Bcl-2/Bax ratio.Conclusion1. Compound angelica injection can inhibit proliferation of myofibroblasts,it had a targeted characteristic.2. Compound angelica injection induces apoptosis in myofibroblasts by up-regulating the bax and down-regulating the bcl-2.3. Compound angelica injection induces apoptosis had an antifibrotic effect in MFB.It may therefore be a candidate for the treatment of OSF.
