Objective:To observe the therapeutic effect of the different doses of AKF-PD on dimethylnitrosamine (DMN)-induced rat liver fibrosis.Methods:234wistar male rats were randomly divided into seven groups, normal group, model group, AKF-PD (15mg/kg,30mg/kg,60mg/kg,120mg/kg) treatment group. PFD (120mg/kg) treatment group (n=30). Model group and treatment group,1%of the DMN intraperitoneal injection,1ml/kg once a day, three consecutive times a week, a total of three weeks; in the third week of the modeling, the treatment group received the corresponding drugs, the corresponding dose gavage once a day, the model group were given0.5%CMC of Na gavage once daily for four weeks; the normal group for each rat to give1ml/kg saline by intraperitoneal injection once a day, three times in a row a week for three weeks were given0.5%CMC Na6ml/kg gavage once daily for four weeks. All rats were sacrificed after four weeks of treatment, specimens from the liver tissue were performed:(1) liver tissue pathology,(2) liver inflammation semi-quantitative score and liver fibrosis semi-quantitative score,(3) immunohistochemical to detect the expression of type Ⅰ、Ⅲ collagen.Results:(1) The influence of AKF-PD on liver inflammation semi-quantitative score:the liver inflammation activity score of DMN model group was significantly higher than the normal group (P<0.05); AKF-PD15mg/kg,60mg/kg treatment groups were significantly lower compared with the model group (P<0.05), the trend is lower of AKF-PD30mg/kg,120mg/kg treatment group than the model group (P>0.05); there is no significant difference between the AKF-PD12Omg/kg treatment group and PFD120mg/kg treatment group (P>0.05);(2) The influence of AKF-PD on the SSS score of liver fibrosis semiquantitative score:SSS score of the DMN model group was significantly increased than normal group (P<0.05); AKF-PD15mg/kg,60mg/kg,120mg/kg treatment groups were significantly lower compared with the model group (P<0.05), the trend is lower of AKF-PD30mg/kg treatment group than the model group (P>0.05); there is significant difference between the AKF-PD120mg/kg treatment group and PFD120mg/kg treatment group (P<0.05);(3) The influence of AKF-PD on the expression of type I、III collagen in rat liver tissue:①The type I collagen:the expression of DMN model group of type I collagen was significantly higher than the normal group (P<0.05); AKF-PD15mg/kg,30mg/kg,60mg/kg,120mg/kg treatment groups were significantly lower compared with the model group (P<0.05); there is significant difference between the AKF-PD120mg/kg treatment group and PFD120mg/kg treatment group (P<0.05);②The type III collagen:the expression of DMN model group of type Ⅲ collagen was significantly higher than the normal group (P <0.05); AKF-PD30mg/kg,120mg/kg treatment groups were significantly lower compared with the model group (P<0.05), the trend is lower of AKF-PD15mg/kg,60mg/kg treatment group than the model group (P>0.05); there is significant difference between the AKF-PD120mg/kg treatment group and PFD120mg/kg treatment group (P <0.05).
