Objective:By detecting the expression of transforming growth factor-β1(TGF-β1) and Connective tissue growth factor (CTGF) in simple benign prostatic hyperplasia(BPH) and BPH combined with imflammation,we attempt to evaluate the effect of inflammation on the development and progression of BPH and to study the relationship between the expression of TGF-β1and CTGF and the clinical parameters of BPH.Method:60prostatic specimens of BPH patients were collected during the operation between August2009and October2011in Haikou Hospital.Recorded the messege of patient’s clinical parameters such as the IPSS、prostate volume (PV)、maximum flow rate(Qmax)、total prostate-specific antigen(tPSA)、postvoid residual urine(PVR).All of60patients were divided into two groups according to suffer from imflammation or not by observing the specimens which were stained with hematoxylin and esin under light microscope. Immunohistochemical method was used in studying the expression of TGF-β1and CTGF.The Iamge Pro-Plus6.0software were used to analyze the MOD(Mean optical density) of each specimen.Statisstical analysises were performed with the use of SPSS13.0software.Two specimens equal numbers were tested by T-test,comparisons among the three groups were made by one-way analysis of variance(ANOVA) and correlative analysis between two variable quantities were analyzed by Pearson-analysis,and for all statistieal tests,the level of significance was set at P<0.05.Results:1.In all of60BPH patients,43were found combined with chronic inflammation,the incidence is71.6%.Low-grade accounting for43.3%,moderate inflammation accounting for18.3%,high-grade accounting for10%.2.There exists a correlation between TGF-β1and CTGF(r=0.836、0.753, P<0.001); The TGF-β1and CTGF MOD in BPH combined with inflammations was higher than that in BPH (P<0.001),but the TGF-β1and CTGF MOD in low-grade、moderate and high-grade inflammation were not significantly different (F=0.178,P=0.837;F=0.035,P=0.965).3.Both of average prostate volume and maximum flow rate(Qmax) of BPH combined with inflammations are higher than the BPH (t=1.999,P=0.048;t=-2.674,P=0.010),but there was no significant difference in IPSS、tPSA、PVR between the two groups (P>0.05);4.MOD of TGF-β1and CTGF,in the group of BPH combined with inflammation,27patients with PV>50ml was higher than the cases whose PV<50ml;38cases of BPH patients of higher IPSS was not significantly different compared with the5cases with the lower IPSS team;19cases of BPH patients whose PVR>50ml was not significantly different compared with the24cases with PVR<50ml;36cases of BPH patients whose Qmax<10ml/s was higher than that in the7cases with Qmax>10ml/s;21cases of BPH patients whose tPSA>4ng/ml was not significantly different compared with the22cases with tPSA<4ng/ml.Conclusion:1. Inflammation is often found in most BPH samples.2. Inflammation can up-regulate their expression in BPH which appeared to play an important role in the development of BPH.3. TGF-β1and CTGF may have a relationship with prostate volume and maximum flow rate and have no correlation with the IPSS、tPSA and postvoid residual urine.
