Comparation Of The Frequency Of Th17Cells In Peripheral Blood In Different Types Of Autoimmune Diabetes

Objective This study aims to investigate the frequency of Thl7cells in T1ADM patients,LADA patients and healthy individuals,and to study the relationships among the frequency of Th17cells, patients’age, the duration of diabetes, HbAlc level,autoantibody titer and islet function. To primarily research the role of Th17cells in the pathogenic machanism of autoimmune diabetes.Research Design and methods The patients with T1ADM (n=10),LADA (n=10) were recruited at Endocrine Department of the Second Xiangya Hospital,Central South University.And age, sex-matched healthy individuals (n=20) were enrolled at the same time. The5ml peripheral blood samples were obtained from40individuals, and then the peripheral blood mononulear cells(PBMC) were separated. The PBMCs were maintained at humidified atomosphere of95%air/5%CO2at37℃for five hours in fresh RPMI1640culture medium supplemented with PMA, ION, MON. After washing, fixation, permeabilization and staining, the frequency of CD3+CD4+IL-17+Thl7cells in peripheral blood were measured by flowcytometry.Results (1) The frequency of Th17cells in T1ADM group and control group was0.62±0.31%vs0.65±0.47%(P>0.05).(2) The frequency of Th17cells in LADA group and control group was 0.64±0.30%vs0.67±0.29%(P>0.05).(3) There is no difference in the frequency of Th17cells between T1ADM and LADA patients.(4) The freqency of Th17cells in both T1ADM and LADA group wasn’t related with the patients’age,duration of diabetes,HbAlc level,autoantibody titer,fasting C-peptides and postprandial C-peptides.But the frequency of Th17cells in T1ADM group was positively correlated with AC-peptides (r=0.891, P<0.01)Conclusions There is no difference in the frequency of Th17cells in peripheral blood between different types of autoimmune diabetes.
