Objective:To research the effects of Icariin on experimental autoimmune thyroiditis(EAT)in the rat model of thyroglobulin antibody(TGAb),thyroid peroxidase antibody(TPOAb)levels,cytokines and the pathological changes of the Thyroid tissue under the light microscope.Methods:30 Wistar rats were divided into five groups randomly,6 rats for each group.They were normal group,the EAT positive control group,low dose group of icariin,middle dose group of icariin and high dose group of icariin.After building the model and the icarrin intervention,ELISA enzyme league immune detected rat serum autoantibodies TGAb and TPOAb levels and cytokines of IL-17?IL-6 and TGF-?.Separated the thyroid tissue,then observed pathological changes of the thyroid tissues and using immunohistochemical method to detect the expression of IL-17.Results:1.Compared with normal group,the EAT positive control group rats serum TGAb,TPOAb level increased significantly(P<0.05);Compared with model group,icariin dose group of TGAb,TPOAb level significantly decreased(P<0.05).2.Compared with normal group,the serum levels of IL-17 in model group significantly increased(P<0.05);Compared with model group,the level of IL-17 in treatment groups were lower,and the middle dose group significantly decreased(P<0.05).Compared with normal group,IL-6 levels have fallen in model group,but no statistical significance(P>0.05),and no significant difference between the treatment group and model group(P>0.05).Compared with normal group,model group's TGF-beta's serum level significantly increased(P<0.05);Compared with model group,the level of TGF-beta treatment group were decreased,the middle dose group was statistically significant(P<0.05).3.Compared with normal group,model group were observed under the thyroid follicular cell damage is better than normal group;Compared with model group,icariin of each treatment group were observed under the thyroid follicular cells were lighter than model group,of which the dose group were observed under the thyroid pathological injury improved obviously.4.Compared with normal group,IL-17 marked the number of cells in thyroid organization in model group is significantly higher than normal group(P<0.05);Compared with model group,each group of the number of IL-17 marked cells were less,especially in medial-dose group(P<0.05).Conclusion:1.Under the proper dosage,icariin in both to reduce the antibody titer and the destruction of the thyroid follicular cells has significant advantages.2.The immunoreaction of Hashimoto thyroiditis is likely by cytokine IL-17,which may be induced by IL-6 and TGF-beta.icariin can not only reduce the levels of IL-17,but also can reduce the level of TGF-beta,double way to suppress the immune reaction.
