The Experimental Research Of Differentiation From Bone Marrow Stromal Stem Cells To Cardiac-like Myocytes By Medicinal Serum Of Xuefuzhuyutang

Objective: To disscuss Xuefuzhuyutang （a medical prescription of promotingblood circulation by removing blood stasis）medicated serum differentiate rat bonemarrow stromal stem cells into cardiac-like myocyte in vitro is feasible.Method: Use medicated serum of Xuefuzhuyutang to induct the Wistar rat bonemarrow stromal stem cells (BMSCs) differentiation in DMEM/F12culture fluid, detectthe BMSCs’ vigor by trypan blue rejection test and OD value by enzyme linkedimmunosorbent assay (ELISA).Then, to observe the cell shape with invert microscope; toidentify the specific markers such as myocardial Desmin, Actin and cTnI withimmunocytochemical staining; to identify myocardial cTnI with lmmunofluorescencemethod and to identify α-MHC and β-MHC of myocardial cell with reverse transcriptasepolymerase chain reaction (RT-PCR).Result: After7days of induction, the BMSCs are wider, and in the shape of a longspindle-shaped, round and oval. Also the BMSCs extend out of the elongatedprotuberances, which appear at the end of the branch, and the orientation is consistent.Portion of the adjacent cells’ protrusions are connected into a network and there aremyotube-like structures4weeks later.,The differentiating cell expressed Desmin, Actinand cTnI when the immunocytochemical staining was performed on them, expressedcTnI under the lmmunofluorescence method, and expressed myocardial cell specificmakers and genes such as α-MHC and β-MHC. Conclusion: The differentiation from bone marrow stromal stem cells tocardiac-likemyocytes by medicinal serum of Xuefuzhuyutang is feasible.
