ObjectiveObjective To evaluate the influence of different concentration of BoneMorphogenetic Protein2(BMP-2) in vitro,cultured chondrogenesis by co-cultureof bone marrow stromal cells (BMSCs).Then search for the optimalconcentration of BMP-2applied in vitrol.MethodsRabbit BMSCs and chondrocytes were in vitrol isolated,cultured andexpand respectively,then mixed up with the ratio of7:3(BMSCs:chondrocytes).The mixed cells were seeded into different culture medium whichadded different combinations and concentrations of growth factor. In thisexperiment, the concentrations of the BMP-2were0ng/ml、5ng/ml、10ng/ml、20ng/ml、30ng/ml、40ng/ml、50ng/ml. We measured the OD of MTT、expressionsof collagen typeⅡand the GAG protein.ResultChondrocytes can provide a chondrogenic microenvironment to induce achondrogenic differentiation of BMSCs in vitro.The gropes which added intoBMP-2showed more GAG formation.ConclusionBMP-2promotes in vitro chondrogenesis by co-culture of BMSCs and asmall amount of chondrocytes. the both factors have synergetic reaction,and the optimal concentration of BMP-2is20ng/ml.
