OBJECTIVES:To investigate the effect and safety of artificial meningeal compounded of BMSCs on repairing infarcted heart.METHODS:BMSCs were obtained by whole bone marrow culture and then subcultured, the growth and the proliferation of BMSCs were observed under inverted phase contrast microscope daily. The third passage of BMSCs were labeled with DAPI and then seeded on artificial meningeal which was sterilized and impregnated with F12-DMEM. The only cell-seeded without artificial meningeal group was cultured in the same conditions as control. To observe the combination of BMSCs and scaffold under an inverted phase contrast microscope and determine the adherence rate. Then the cell proliferation was detected by MTT. MI in Sprague-Dawley (SD) rat was established by ligation of the left anterior descending coronary artery.60Rats were randomly divided into four groups:sham-operated, MI, MI+cell-free artificial meningeal, and MI+BMSC-seeded artificial meningeal. The sham group just underwent the identical surgery but without ligation of the coronary artery. MI+cell-free artificial meningeal animals received the MI operation and the transplantation of cell-free artificial meningeal. MI+BMSC-seeded artificial meningeal transplanted a BMSC-seeded artificial meningeal which was cultured for3days on the surface of ischemic and peri-ischemic myocardium after MI. Four weeks after MI operation and patch transplantation, cardiac function were evaluated in four groups of rat, and the expressions of Cx43protein were detected by Western blot, the survival rate were caculated at the same time.RESULTS:The adherence rate and the cell proliferation represented a time increasing trend, and there were no significant differences compared with the control group at different time points(P<0.05). Compared with MI alone and MI+cell-free artificial meningeal groups, the rats received MI+BMSC-seeded artificial meningeal significantly improved left ventricular function, the expressions of Cx43protein and the survival rate increased obviously(P<0.05).CONCLUSION:BMSCs adhesioned, grew and proliferated well on the artificial meningeal. Artificial meningeal with BMSCs can improve cardiac function and survival rate in MI rats.
