Acanthopanax senticosus (Acanthopanax senticosus(Rupr.et Maxim)Harms) also known as Acanthopanax Root which is the dry roots,rhizomes and stems of the Araliaceous. Acanthopanax senticosus has the effects on tonifying Qi and strengthening the spleen,tonifying kidney and tranquilization,nourishing essence, strengthening the will and physique. It is good for symptoms such as Yang deficiency of spleen and kidney, body weakness, loss of appetite, waist and knee pain, insomnia and dreamfuletc.Extractum Acanthopanacis Senticosi is extract of water or ethanol of Acanthopanax senticosus, in which Isofraxidin, Syringin and Eleutheroside E are the specific component of Extractum Acanthopanacis Senticosi, with a variety of pharmacological effect. Isofraxidin is the main component functioning on calming,sedative effects Acanthopanax senticosus.Syringin functions against the fatigue and hemostasia.Isofraxidin, Syringin and Eleutheroside E are the components of the evaluation of quality indicators.Under the item of [Extractum Acanthopanacis Senticosi] and[Content Determination] in Chinese Pharmacopoeia (2010Edition)[1],3components are determined in the same detection wavelength (220nm) and the same flow chromatography conditions.We believe that as the structures of the three are different,the maximum absorption peak wavelength is also different.Choose their self absorption peak wavelength to determine is much consistent with the real content,moreover,220nm as the end of ultraviolet of the determination effect is not ideal. Based on this,the experiment carry out the test by using different chromatographic conditions to measure in the extract of isofraxidin,syringin content determination (Limited to the conditions we haven’t studied the component of Eleutheroside E)..At the same time,Enrichment method of the main target components of Extractum Acanthopanacis Senticosi in the [Pharmacopoeia] is studied.In research of the Content determination, at first we investigated method to extract the Isofraxidin and syringin in Extractum Acanthopanacis Senticosi.Different concentrations of ethanol, extraction time and different extraction times were firstly investigated by orthogonal test method,then we make sure the optimum extraction scheme that is with75%methanol to extract three times,each time1.5hours.Secondly,this study using the method of HPLC,investigate the precision,stability,repeatability,sample recovery rate experiment research on content determination method of Isofraxidin and syringin in Extractum Acanthopanacis Senticosi respectively.The result of Experiment of Isofraxidin show that under the wavelength of344nm and within0.016-0.16ug/ul limits,Isofraxidin has a good linear relation and its correlation coefficient is0.9999(n=5); Syringin under wavelength of365nm,and within the0.075-1.5ug/ul limits,has a good linear relationship, and its correlation coefficient is0.9993(n=5).Determination of the three batches of Acanthopanax extract samples,the content of Isofraxidin is average of0.11percent; Syringin is0.68%. The experimental results show that the method is simple and reproducible and provide reliable quantitative basis for the control of the quality of Isofraxidin and Syringin in Acanthopanax senticosus.This experiment also examines the process of enrichment the content of Isofraxidin and syringin in Extractum Acanthopanacis Senticosi with5types of macroporous resins like AB-8.Investigated the leaking curve,the best time of adsorption, elution velocity and elution solvent.Considerate comprehensively, the AB-8type resin is the first choice.The best technology:by computed the maximum sample volume is0.26g Extractum Acanthopanacis Senticosi in1ml resin.Under these conditions, the content of Isofraxidin and Syringin in Enrichment is respectively4.2and4.3times.
