Objective: To study the expression of NF-κB in hypertensive intracerebralhemorrhage(HICH) secondary brain injury.Methods:30cases of hypertensive intracerebral hemorrhage patients werecollected in neurosurgery of the second affiliated hospital of nanhua universitybetween the October in2010and the April in2011.Basal ganglia hemorrhage,hematoma without involvement of cortical. Took the cortical brain tissue as controlgroup. Took the basal ganglia of perihematomal brain tissue as experimental group.Specimens were divided into male group and female group on the base of gender.Specimens were divided into graded III and graded IV group on the base of Cerebralhemorrhage consciousness status classification. Using Annexinâ…¤-FITC/PI doubledyeing, detecting apoptotic cells proportion with flow cytometry instrument TheEnzyme-Link ImmunoSorbent Assay method and immunohistochemistry methodwere used to determine the expression of NF-κB proein in all specimens.Results:1〠Flow cytometry instrument detect the percentage of apoptotic: cellapoptosis phenomenon are detected all of specimens. Experimental groupcompared with control group, the differences has statistically significant.(P=0.00,P<0.05).2ã€Enzyme-Link ImmunoSorbent Assay:NF-κB are detected all of specimens.Experimental group compared with control group, the differences has statisticallysignificant(P=0.00,P<0.05).3ã€Enzyme-Link ImmunoSorbent Assay: NF-κB in the hematoma of â…£group compared with III group, the differences has statistically significant(P=0.036,P<0.05). 4ã€Enzyme-Link ImmunoSorbent Assay: NF-κB in the hematoma of malegroup compared with female group, the differences has statistically significant(P=0.036,P<0.05).5ã€Immunohistochemical:The NF-κB positive cells are visible in the aroundthe hematoma and normal brain tissue groups. The positive cells in the around thehematoma group is (21.33±3.21),the normal brain tissue group(7.67±1.53)The positive percentage of NF-κB in the around the hematoma group were higherthan the normal brain tissue group (P=0.014,P <0.05).Conclusions:1ã€The cell apoptosis phenomenon appears in the early of hypertensiveintracerebral hemorrhage. The percentage of apoptosis around the hematoma is higherthan cortical brain tissue.2ã€NF-κB exist in the early of hypertensive intracerebral hemorrhage. NF-κB around the hematoma is higher than cortical brain tissue.3〠The expression of NF-κB in hypertensive intracerebral hemorrhage hasnot related with gender.4ã€The expression of NF-κB in hypertensive intracerebral hemorrhage hasrelated with Cerebral hemorrhage consciousness status classification.5ã€The expression of NF-κB may related with the brain cell apoptosis afterencephalorrhagia.6ã€The effection of NF-κB in hypertensive intracerebral hemorrhage stillneeds further study. |
