[Objective] This study was designed to explore the effect of Salvia yunnanensis C. H.Wright on the fibroblast of scar and the pig back scar model about its biological behavior and its pathological changes. Reveal the mechanism to provide basic theory for clinical precaution and treatment to scars at molecular and pathological level.[Methods]l. The fibroblasts of scars were cultivated in culture fluid containing different concentration Salvia yunnanensis C.H.Wright (0.05g/ml,0.1g/ml,0. 2g/ml,0.4g/ml). The results were observed and analyzed by inverted microscope, MTT assay, immunocytochemistrical techniques and Western blot. Above techniques are used to analyzed cell cycle and the content of decorin. 2. Hypertrophic scar models were made on pig back in vivo by different concentration Salvia yunnanensis C.H.Wright injection, The content and distribution of decorin in the tissue samples were determined with immunohistochemistry. Using these experimental analyse, to study the biological behavior and pathological changes of hypertrophic scar fibroblasts affected by Salvia yunnanensis C.H.Wright.[Results] 1. In vitro: compared with the control group, by LM, the density of cells depressed and the spindle-shape cell were discovered becoming retractation and abnormity. MTT assays showed significant difference between the two groups (P<0. 05). By immunocytochemistrical techniques and Western blot: compared with control groups, the decorin content was increased in the experiment group.2. In vivo (pig back scar model): The expression of decorin was detected by immunohistochemistry: The expression of decorin was rich in the normal skin dermis. The lower expression of decorin was found after two weeks with Salvia yunnanensis C.H. Wright injection, but the control group were not found. [Conclusions] Salvia yunnanensis C. H. Wright can decrease fibroblasts growth, proliferation and collagen synthesis and increase the content of decorin.
