Objective:To prepare rat model which can simulate pediatric cough variantasthma,combined with modern medical related research,using modern microscopicand immune molecular biology technology, by means of measuring and analyzingdifferent grouping rats lung tissue pathological,serum IL-4,IFN-γ content andIFN-γ/IL-4,observe the effects of Ke Sou particles flavored on lung tissue structureand immune function of cough variant asthma in rat models and then discusses the KeSou particles adjust lung tissue pathology and immune dysfunction mechanism ofcough variant asthma,provide powerful laboratory evidence for clinical medication.Methods:according to the method of cough variant asthma in guinea pigs model,copy cough variant asthma in the rat model.60wistar rat healthy are randomly dividedinto six groups of10only. Groups A:normal control group;Group B:CVA modelgroup;Group C:Xiaoerfeirekechuan particles group;Group D:Ke Sou particles lowdoses groups;Group E:Ke Sou particles middle-dosage groups;F:Ke Sou particleshigh-dosage groups.To start experiment after normally breeding3days,in addition tonormal control group,the first,8th days every rats of the rest in each group hasintraperitoneal injection10%egg albumin and AL (OH)3mixed solution1mlsensitization.From the15th day,B,C,D,E,Frats,placed in airtight organiccontainer,given the physiological saline of1%OVA to ultrasonic atomizer stimulate20minutes, once a day, a total of10days, arouse before30minutes,C, D, E, F groupswere gastric lavage to medication.The26th day,cough times determination,theabdominal aorta took blood and separate serum IFN-γ,IL-4cytokines level measuring,executed lung tissues of rats did return on pathological examination.Results:1Compared with normal group,the cough number of model groupsignificantly increased,has significant difference(P<0.05);compared with the CVAmodel group,Xiaoerfeirekechuan particles group,Ke Sou particles each dosage grouphas diminished, there is significant difference(P<0.05);and each dose group of Ke Sou particle relatively is less compared with Xiaoerfeirekechuan particles group,hassignificant difference(P<0.05);compared with high dose group,the cough times oflow,medium dose group is relatively much,has significant difference(P<0.05).2Compared with normal group,the serum IFN-γ of CVA model group significantlyreduces,with significant difference(P<0.05);compared with model group,the serumIFN-γ of Xiaoerfeirekechuan particles group,Ke Sou particles each dosage groupsignificantly increases,with significant difference(P<0.05);and the sereum IFN-γ ofeach dose group of Kou Sou paticles is relatively increased compared withXiaoerfeirekechuan particles group,with significant difference(P<0.05),thehigh-dosage groups has significant difference compared with low,medium dosegroup(P<0.05).3Compared with normal group,the serum IL-4of CVA model groupsignificantly increases,with significant difference(P<0.05);compared with modelgroup,the serum IL-4of Xiaoerfeirekechuan particles group,Ke Sou particles eachdosage group significantly reduces,with significant difference(P<0.05);and thesereum IL-4of each dose group of Kou Sou paticles is relatively reduced comparedwith Xiaoerfeirekechuan particles group,with significant difference(P<0.05),thehigh-dosage groups has significant difference compared with low,medium dosegroup(P<0.05).4Compared with normal group,all levels within the bronchial wallmucosal epithelium damage degree,mucus plugs form proportion andsize,inflammatory cell and the surrounding alveolar involvement etc pathologicalchanges of CVA model group are obviously serious;Compared with model group,thepathological changes of Xiaoerfeirekechuan particles group,high, medium and lowdose group of Ke Sou particles significantly reduce; Ke Sou particles dose group ismilder than Xiaoerfeirekechuan particles group;the high-dosage groups is milder thanlow,medium dose group.Conclusion:1Ke Sou particles can obviously reduce the degree of airwayinflammatory response.2Ke Sou paticles can significantly improve the IFN-γ level ofserum in CVA rats,reduce the serum IL-4content of rat models, have obvious effect onadjusting the IFN-γ/IL-4immune disorders.
