Objective: To quantify the expression of FXYD3in the human colon carcinomatissue, and to investigate the significance of relationship between the expression of FXYD3protein and clinicopathological parameters in the patients with colon carcinoma.Methods: Protein levels of FXYD3in paraffin sections of60cases of coloncarcinoma,34cases of adjacent normal mucosa and20cases of distant normal mucosawere determined by Immunohistochemical Streptavidin-perxiodase technique, and mRNAlevels of FXYD3of13cases of colon carcinoma and distant normal mucosa wasquantified by RT-PCR. Additionally, clinical parameters including gender and age of thesepatients and the site, grade were recorded, and the protein expression levels of p21, p53,MDR, ki-67of colon carcinoma were determined by immunohistochemistry. Finally, therelationship between the expression of FXYD3protein levels and clinicopathologicalparameters in the patients with colon carcinoma were analyzed by Chi-square test andWilcoxon rank sum test.Results: Immunohistochemical results showed that FXYD3protein was expressed inboth of cell membrane and cytoplasma. The positive rate of FXYD3protein expression indistant normal mucosa and adjacent normal mucosa were100%and85.29%, which weresignificantly higher than that in colon carcinoma of50%(P<0.05). The expression ofFXYD3mRNA in colon carcinoma were lower than that in distant normal mucosa. Theexpression of FXYD3protein was positively correlated with FXYD3mRNA in the samecolon carcinoma, but this was not statistically significant (P>0.05).There was a closerelation between lower FXYD3expression in colon carcinoma and higher pathologic grade(P<0.01), later clinic stage (P<0.01), lymph node metastases (P<0.01). The expressionof FXYD3protein was positively correlated with p21, MDR, but this was not statisticallysignificant (P>0.05). There was a positive association between FXYD3protein and p53 (r=0.29446, P<0.05), whereas a negative association between FXYD3protein and ki-67(r=-0.30376, P<0.05).Conclusion: The FXYD3proteins and mRNA in colon carcinoma were lower thanthose in distant normal mucosa. Lower FXYD3expression may be positively correlatedwith the development of the relative later stages of colon carcinoma. The expression ofFXYD3protein and p53, ki-67was correlated in colon carcinoma.
