OBJECTIVE : To detect the expression of leucine-rich repeat-containing G-protein-coupled receptor 5 and stromal cell-derivedfactor-1 in colon carcinoma and the clinical significance and to investigate the relationship between LGR5 and SDF-1 expression and biological behavior of colon carcinoma.METHOD:The expression of LGR5 and SDF-1 in 80 cases of colon carcinoma and 20 cases of normal colon epithelium were detected by immuhistochemistry method. Analyze the expression of LGR5 and SDF-1 compared with the clinical stage, pathological grade and distant metastasis of colon carcinoma patients.RESULT:1. Expression of LGR5 and the correlation with the clinicopathological factors① The positive rate of LGR5 in normal colon epithelium and colon carcinoma were 5.00%(1/20) and 55.00%(44/80)respectively.The difference was significant between every two of them(P<0.01).② The positive rate of LGR5 in clinical stage>ⅡC(65.30%)was obvious higher than the stage ≤ⅡC(38.70%),(P<0.01);In addition, the LGR5 positive rate in cases with distant metastasis was significantly higher than the cases without distant metastasis.(P<0.01); The LGR5 positive rate in pathological grad Ⅲ was not significantly higher than the grade Ⅰ+Ⅱ cases.( P>0.05)。③ The positive rate of LGR5 in colon carcinoma had no relation with patient’s age and gender.(P>0.05,P>0.05)2. The relationship between expression of SDF-1 and its clinicopathological factor in colon carcinoma.① The positive rate of SDF-1 in normal colon epithelium and colon carcinoma were 15.00%(3/20)and 61.25%(49/80) respectively. The SDF-1 positive rate in colon carcinoma was significant higher than normal colon epithelium cases.( P<0.01)。② The positive rate of SDF-1 in clinicopathologic stage >ⅡC(73.47%)was clear higher than the stage ≤ⅡC(41.93%)( P<0.01); The SDF-1 positive rate in cases with distant metastasis was significantly higher than the cases without distant metastasis( P < 0.05); The SDF-1 positive rate in pathological grad Ⅲ was significantly higher than the grade Ⅰ+Ⅱ cases.(P<0.05)。③ The positive rate of SDF-1 in colon carcinoma had no relation with patient’s age and gender.(P>0.05,P>0.05)。3. Relationship between expression of LGR5 and SDF-1The expression of LGR5 had a positive correlation with the expression of SDF-1. The higer the expression of LGR5, the higher the expression of SDF-1.(R=0.260,χ2=5.43,0.01<P<0.05)。CONCLUSION:The higher expression level of LGR5 and SDF-1 are involved in the colon carcinoma cells proliferation, malignant transformation and metastasis. They may both play an important role during the occurrence and development of human colon carcinoma.
