Objective To reproduce the rats models with incisional pain, compare the analgesia effect of flurbiprofen axetil in rats by giving different doses of flurbiprofen axetil at two time points, discuss the preemptive analgesia effect of flurbiprofen axetil.(1) To observe the rats behavior research and COX-2of spinal cord by injection different doses of flurbiprofen axetil from the tail vein on the rats models of incisional pain.(2) To research the dose dependent by injection different doses of flurbiprofen axetil from the tail vein.Methods72male Wistar rats were randomly divided into3groups:the control group(E group),preoperative treatment group(P group)and postoperative treatment group(C group).P group and C group were divided into4groups respectively, which were saline(P1,C1group), flurbiprofen axetil1,3,9mg/kg group(P2,P3,P4group,C2,C3,C4group). E group(n=8)without operation but anesthetized only, the rats of P group and C group were made according to the Brennan method, P group received saline or flurbiprofen axetil before incision and C group15minutes after incision through tail vein. After rats came back to consciousness and1hour surgery, use cumulative pain score test to evaluate the pain behavior, and2hours later, kill all the rats, remove the spinal cord and examine the COX-2expression of spinal cords by ELISA.Results (1)E group:the rats without operation does not have the change of behavior, compared with E group, cumulative pain scores and COX-2expression of spinal cord of P and C group were higher, there is significant difference(P<0.05);(2)P group:Compared with P1group, cumulative pain scores and COX-2expression of spinal cord of P2,P3and P4groups were lower, there is significant difference(P<0.05);(3)C group:Compared with C1group, cumulative pain scores and COX-2expression of spinal cord of C2,C3and C4groups were lower, there is significant difference(P<0.05);(4)the comparison of P group and C group:there is no significant difference(P>0.05)between P1group and C1group,P2group and C2group,P3group and C4group, while P3and C3group,P4and C4group difference was significant(P<0.05),cumulative pain scores and COX-2expression of spinal cord of P4group is the lowest.Conclusion Flurbiprofen axetil can suppress the pain reaction of rats with incisional pain impactful,COX-2expression of spinal cords were suppressed by flurbiprofen axetil in dose dependent, flurbiprofen axetil has showed preemptive analgesia effect.
