Objective Thyroid associated ophthalmopathy (Thyroid Associated Ophthalmopathy TAO),the one of the most common orbital diseases in the adult,is a autoimmune disease.TAO is basedon the characteristics of the eye and per orbital tissue invasive disease and closely related togenetic factors. Peroxisome proliferator activated receptor gamma (PPAR gamma) is one of thedifferentiated transcriptive factors in the adipose differentiation process, and experimentalstudies have shown that TAO patients retrobulbar fat or connective tissue owns significantlyhigher gene level in PPARγgene expression level than normal control group, and the increasein PPARγgene expression may be one of the pathogenic factors in TAO. This experimentthrough the study of PPAR ofγpro12ala polymorphism in the distribution of part of the Hanpeople in Shanxi Province and the association with thyroid associated ophthalmopathy (TAO),in order to explore the genetic factors and pathogenesis of TAO and provide some experimentalevidence for diagnosis.Methods Subjects a total of 166 cases divided into three groups, of which 51 cases of TAOpatients, 55 patients of Graves’ disease (GD) and normal controls was 60 cases. Test PPARγpro12ala genotype and allele frequency in Shanxi Han people by PCR RFLP method. Thenusing the application SPSS17.0 statistical software to analyze the individual sample allele andgenotype. Allele and genotype frequencies in the groups were compared using chi square test.Results G allele in TAO group frequency (27.8%) was significantly higher than that of GDpatients or normal control group (13.6%, 13.3%) (P <0.05); CG + GG genotype frequency inTAO patients group (52.9%) was significantly higher than that of GD patients and normalcontrol group (25.5% 26 7%) (P <0.05), the GD group and normal control group, allele and genefrequency difference was not statistically significant.Conclusion(1)There is PPARγgene polymorphism pro12ala in Chinese Han people in ShanxiProvince;(2)PPARγgene polymorphism pro12ala may be associated with the incidence ofTAO in Chinese Han people in Shanxi Province.
