Objective:Using Quantitative real time polymerase chain reaction (qRT-PCR) to detected expression of the microRNA-29b from formalin-fixed paraffin-embedded(FFPE) tissue blocks of endometrium,to explore its clinicai significance during carcinogenesis of endometrium.Methods:Choosing96cases endometrium of formalin-fixed paraffin-embedded(FFPE) tissue blocks(including16normal cases(secretory phase), atypical hyperplasia20cases and endometrial carcinoma60cases),according to the sequence of microRNA-29b and U6,the stem-loop reverse transcriptional primers and the polymerase chain reaction primers were designed,expression of the microRNA-29b during carcinogenesis of endometrium were detected by Quantitative real time polymerase chain reaction estrogen receptor(ER),progesterone receptor(PR) were detected with immunohistochemistry. The association of the microRNA-29b expression with the clinicopathologic features of endo-metrial carcinoma and estrogen receptor(ER),progesterone receptor(PR) were analyzed.Results:(1)The protein positive rates (70%,56.67%) of estrogen receptor(ER),progesterone receptor(PR) in endometrial carcinoma were lower than that in normal endometrium and atypical hyperplasia endometrium (100%,93.75%;85%,90%)(P<0.05).(2) Expression levels of the microRNA-29b in endometrial carcinoma(0.825±0.222) were lower than that in normal endometrium and atypical hyperplasia endometrium (1.013±0.160,0.949±0.163)(P<0.05).(3)Expression levels of the microRNA-29b in clinical stage III+IV (0.730±0.212)were lower than that in clinical stage Ⅰ+Ⅱ (0.860±0.217)in endometrial carcinoma;Expression levels of the microRNA-29b in presence of lymph node metastasis (0.633±0.136) were lower than that in none of lymph node metastasis (P<0.05).(4) Expression levels of the microRNA-29b in negative expression of estrogen receptor(ER),progesterone receptor(PR) compared with positive expression were not significant(P>0.05).Conclusion:microRNA-29b probably has a relationship with the occurrence and development of endometrial carcinoma,microRNA-29b may be helpful for diagnosis of endometrial carcinoma.
