Objective:Antibacterial and antiviral experiment are to explore the inhibitory action of Zhebeizhike granules on pathogenic microorganisms and protective mechanisms of Zhebeizhike granules for infected mice.The pharmacodynamics studies are to confirm the effectiveness of Zhebeizhike granules on influenza viral H1N1infected mice and the antitussive and expectorant effects of Zhebeizhike granules.Methods:Experimental research on antibacterial action of Zhebeizhike granules was applied to observe the antibacterial effect. Cell survival rate and inhibition rate were served as target to evaluate antiviral activity of HPE against influenza viral H1N1in vitro by method thiazolyl tetrazolium(MTT)and the influence of Zhebeizhike granules against proliferation of influenza viral H1N1in MDCK cell.Mice were infected with influenza viral H1N1through nasal cavity. And than infected mice were treated with Zhebeizhike granules by mouth for7days,The major criteria for evaluation including influence on the mortality rate, lung index and the change of viral pneumonia. Pathomorphology of the lung tissue of mice were observed to approach further the mechanism of Zhebeizhike granules on mice infected with influenza viral H1N1. The antitussive effect was studied by the cough model induced by ammonia water in mice, the expectorant effect was studied by a mouse model of phenol red test; the expectorant effect was studied by testing the amount of phlegm secreted in rats.Results:1The in vitro antimicrobial experiment: inhibition zone diameter on Staphylococcus aureus of Zhebeizhike granules is14mm, MIC is10.55mg/mL, MBC is21.1mg/mL; inhibition zone diameter on Streptococcus pneumoniae of Zhebeizhike granules is16mm, MIC is42.19mg/mL,MBC is84.38mg/mL;inhibition zone diameter on alpha hemolytic streptococcus of Zhebeizhike granules is18mm, MIC is42.19mg/mL,MBC is84.38mg/mL;inhibition zone diameter on hemolytic streptococcus of Zhebeizhike granules is20mm, MIC is21.1 mg/mL,MBC is84.38mg/mL.2The in vivo antimicrobial experiment:the mortality in mice of high and middle dose group of Zhebeizhike granules were30%and40%, compared with the model group (mortality100%), the difference was significant(P<0.01). Compared with the positive control, the differences is no significant(P>0.05).3The in vitro anti-virus experiment:TC5o on MDCK of Zhebeizhike granules is110.81mg/mL, IC50is1.41mg/mL, TI is78.6.4The in vivo anti-virus experiment: mice death protection rate of the high and middle dose group of Zhebeizhike granules on infected mice is55.56%, compared with the model group, the difference was significant (P<0.05), Compared with positive control group, there is no significant differences (P<0.05);compared with the model group, the lung index of the high and middle dose group of Zhebeizhike granules were significantly lower(P<0.05); pneumonia lesions of the high and middle dose group of Zhebeizhike granules significantly reduced.5Pharmacodynamics experiment results:compared with the model group, Zhebeizhike granules can significantly extend incubation period of mice cough, reduced cough number (P<0.01), increase the phenol red shedding quantity of mice(P<0.05orP<0.01).Conclusion:1Zhebeizhike granules have inhibitory action to Staphylococcus aureus, Streptococcus pneumoniae, alpha-hemolytic streptococcus and hemolytic streptococcus, and have a protective effect on the mouse death.2Zhebeizhike granules can effectively inhibit replication of influenza virus H1N1in cultured MDCK and CEF, and can improve the symptoms of pneumonia in mice caused by influenza virus, to extend the mice infected with influenza virus survival, reduce mortality, and therefore antiviral drug may be a value drug.3Zhebeizhike granules have obvious antitussive and expectorant actions.
