Objective To investigate the protein and mRNA expressions of Wnt-induced secreted protein-1(WISP-1) and matrix metalloproteinase-2(MMP-2) in colorectal adenocarcinoma, adenoma and normal colorectal tissue. To discuss the role of WISP-1, MMP-2in development of colorectal tumor, and the relationship between the expression of WISP-1, MMP-2and clinicopathologic features of colorectal tumor.Methods(1)The immunohistochemical PV-9000method was used to detect the expressions of WISP-1and MMP-2proteins in112cases of colorectal adenocarcinoma and56cases of adenoma,30cases of normal colorectal tissues as the control groups.(2)Semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) was performed to detect the expression of WISP-1mRNA and MMP-2mRNA in40cases of colorectal adenocarcinoma,12cases of adenoma and40cases of normal colorectal tissues as the control groups.Results (1) The expressions of WISP-1and MMP-2in colorectal adenocarcinoma were remarkably higher than that of in adenoma and normal colorectal tissue(P<0.05). In the cases of colorectal adenocarcinoma, the expression rates of WISP-1and MMP-2were related to the TNM stages, tumor size, lymph node metastasis and serosa invasion (P<0.05). The expression of WISP-1was positively associated with that of MMP-2protein (r=0.732, P<0.05).(2) The expressions of WISP-1and MMP-2in adenoma were remarkably higher than that of in normal colorectal tissue(P<0.05). The expression rates of WISP-1and MMP-2were correlated with histological shape and degree of dysplasia in colorectal adenoma(P<0.05).(3) The WISP-1and MMP-2protein expressions were in accordance with mRNA expression level. From normal colorectal tissue to adenocarcinoma, the expressions of WISP-1and MMP-2were gradually enhanced.Conclusion (1)The aberrant expressions of WISP-1and MMP-2in colorectal adenoma are closely correlation with malignant transformation. They are an early event in the development of adenocarcinoma, and are expected to help the early diagnosis of colorectal adenocarcinoma.(2)The expression rates of WISP-1and MMP-2were related to the TNM stages, tumor size, lymph node metastasis and serosa invasion of colorectal adenocarcinoma. They can be used as a potential indicator to forecast the invasion and metastasis of colorectal adenocarcinoma. Therefore, combined detection of the expressions of WISP-1and MMP-2might be helpful in the evaluation of the prognostic factors in patients with colorectal adenocarcinoma.
