Objective:①The decalcification model of physiological root resorption of human primary teeth will be constructed to provide the experiment conditions on the production of protein molecules②To investigate the function and the expression of RANKL and TRAF6during the stage of physiological resorption of human deciduous teeth in order to add some new evidence for the research.Methods:①Collected the Physiological root resorption of human Primary teeth and divided them into four groups:the early Primary tooth root resorption, the medium Primary root resorption group,the end of Primary tooth root resorption group and the Permanent teeth group, take the orthodontic needing extraetion normal Permanent as the control group. Immediately after extraction, the teeth were fixed by immersion in a mixture of4%formaldehyde. They were then decalcified in15%EDTA solution in order to construct the decalcification model of physiological root resorption of human primary teeth.①Tissue sections of5μm thickness were cut in the sagittal plane. Detected the RANKL and TRAF6expressions of the primary teeth root resorption in Immunohistoehemistry. Using BI-2000image analysis soft ware on each set of experiments sections stained odontoclasts,odontoblasts,pulp fibroblasts accumulated optical density value (integral optical density,IOD) measured.Using One-way ANOVA were statistically analyzed.Results:Immunohistochemistry resultsThe First ExperimentFibroblasts,odontoblasts and odontoclasts accumulated more optical density of RANKL in the experimental groups. There is no optical density of RANKL in the control group. There was significant statistical difference(P<0.01). Root resorption of the medium group was higher than the early and the late groups,there was a significant difference(p<0.01)-odontoclasts accumulated more optical density of RANKL in the same experimental group compared with Fibroblasts and odontoblasts. There is no difference between odontoblasts and fibroblasts; Control group: Expression of RANKL in the odontoblasts and pulp fibroblasts is negative(-),There is no Odontoclasts in the control group.The Second ExperimentFibroblasts,odontoblasts and odontoclasts accumulated more optical density of RANKL in the experimental groups. There is no optical density of TRAF6in the control group. There was significant statistical difference(P<0.01). Root resorption of the medium group was higher than the early and the late groups,there was a significant difference(p<0.01)·odontoclasts accumulated more optical density of TRAF6in the same experimental group compared with Fibroblasts and odontoblasts. There is no difference between odontoblasts and fibroblasts; Control group: Expression of TRAF6in the odontoblasts and pulp fibroblasts is positive(+),There is no odontoclasts in the control group.Conclusions:1.the highest optical density of RANKL in odontoclast was in the middle group of the Physiological root Resorption of Deciduous teeth. Suggesting that the funntion of RANKL is strongest in middle group. The most active function of odontoclasts was during the middle group.2.In the immunohistochemical experiment,the highest optical density of TRAF6in odontoclast was in middle group of the Physiological root Resorption of Primary tooth. Suggesting that the funntion of TRAF6was strongest in middle group.3.The expression of TRAF6in odontoclasts odontoblasts and fibroblasts is consistent with that of RANKL during the experimental groups4.There is no expression of RANKL and TRAF6in both odontoblasts and pulp fibroblasts dring the control group.
