Objective to observe the change of general condition, the expression of heme-oxygenase-1,the content of MDA, concentration of total protein in bronchoalveolar lavage fluid, morphology of lung in sepsis shock rats and explore the possible action mechanism of ketamineMethods40male vista rats were randomly divided into sham group (group S), CLP group (group C), ketamine group(group K), ketamine plus zinc protoporphyrin IX group (group KZ),zinc protoporphyrin IX group(group Z). SHAM group received laparotomy without any treatment, other groups underwent CLP group. Rats were administrated saline lml in group S and C, ketamine40mg/kg in group K, ketamine40mg/kg plus zinc protoporphyrin IX5mg/kg in group KZ, zinc protoporphyrin IX5mg/kg in group Z.All the drugs were diluted to1ml with saline. Rats were sacrificed at the9th h after CLP. The levels of MDA in lung tissue were measured, bronchoalveolar lavage fluid (BALF) was collected for measurement of concentration of total protein. HE staining was used to observe pathological changes of lung. And the level of HO-1was measured by immunohistochemical method.Results①General situation:Compared with the sham group, the activity of CLP group animal decreased, lethargy, piloerection, angle of eyes have Exudates and so on. The general situation of SHAM group was better.②MDA:Compared with group S, the contents of MDA were increased in other injury groups(p<0.01);compared with group C, the contents of MDA were decreased in group K(p<0.01);compared with group K, the contents of MDA were increased in group KZ(p<0.01);compared with group C, there is no significant difference in the contents of MDA in group KZ, Z.③Compared with group S, the contents of TP were increased in other injury groups(p<0.01);compared with group C, the contents of TP were decreased in group K(p<0.05);compared with group K, the contents of TP were increased in group KZ(p<0.01);compared with group C, there is no significant difference in the contents of TP in group KZ, Z.④Compared with group S, the expression of HO-1were increased in other injury groups (p<0.01); compared with group C, the expression of HO-1were up-regulated in group K(p<0.01); compared with group K, the expression of HO-1were down-regulate in group KZ(p<0.01); compared with group C, there is no significant difference in the expression of HO-1in group KA, Z.Conclusion the content of MDA, TP were increased in sepsis rats lungs. Ketamine extenuate the content of MDA, TP in sepsis rats lung through up-regulating HO-1expression. using HO-1inhibitor znpp, the protective effect was decreased.
