Objective: to study the expression and significance of HO-1 in lung tissue of rats in sepsis.Methods: 48 SD rats were randomly divided into two groups, the sham group and sepsis group. Sepsis group 24, Cecal ligation and puncture(CLP) method was used to establish the model of sepsis; sham group 24, only to do switch abdominal processing. Rats from two groups were killed and specimens were taken from the lung tissue, at 6h, 12 h, 24 h. HE staining was done to show the pathological changes in these two groups. HO-1 protein expression was shown by the application of Western blot. HO-1 m RNA was detected through Realtime PCR.Results: 1. from HE staining, the lung tissue of rats showed different pathological changes in different detecting time in sepsis group, the lung tissues in CLP groups showed more obvious hemorrhage, tissue swelling and inflammatory cell infiltration compared with the sham group; 2. By the western blot analysis of HO-1 protein expression, proteins at each time point of sepsis group were significantly higher than those of sham group; 3. Compared with the sham group, HO1 m RNA level significantly increased at 6h, 12 h, 24 h compared with the sham group.Conclusion: 1. In sepsis group, the expression of HO-1 in lung tissue of rats showed difference with the sham group; 2. High expression of HO-1 in sepsis group possibly indicated that HO-1 was involved in lung injury.
