Objective Compare the cumulative pain scores and the content of COX-2and TNF-a of parecoxib sodium on the rats with incision pain by giving different doses of parecoxib sodium at two different time point to investigate the preemptive analgesia effect of parecoxib sodium.Methods54male Wistar rats were randomly divided into three parts:control group (K part),the preoperative group (F part),the postoperative group (H part). Control part(n=8):on ly injected saline.F part:injected saline or parecoxib sodium30min before the operation.H part:injected saline or parecoxib sodium30min after the operation.F part and H part we re further divided into four groups (n=8):the saline group(F1,Hlgroup),parecoxib sodium3,6,12mg/kg groups(F2,F3,F4,H2,H3,H4group).observe the cumulative pain scores within1h after the operation, removing the spinal cord enlargement4h after the surgery,measure th-e content of COX-2in every group and TNF-α in K1,F1,F4,H4groups by ELISA.Results Compared with K part,the cumulative pain scores and the content of COX-2in spinal cord was significantly high in F1and H1groups(P<0.05).In F part, compared with F1group,the cumulative pain scores and the content of COX-2in F2,F3,F4groups decrease(P<0.05),when the dose become larger,the cumulative pain scores and the content of COX-2were lower(P<0.05).In H part,compared with H1group, the cumulative pain scores and the content of COX-2in H2, H3, H4groups decrease(P<0.05), when the dose become larger,the cumulative pain scores and the content of COX-2were lower(P<0.05).The comparison between F part and H part,there was no significant difference between F2group and H2group(P>0.05), while there was significant difference between F3group and H3group,F4group and H4group,F3and F4was lower(P<0.05);on the content of COX-2,there was no significant difference between F2group and H2group(P>0.05),F3group and H3group,while F4group was lower than H4group (p<0.05).On the content of TNF-a in spinal cord,compared with K part,the content of TNF-a was significantly higher in F1,F4,H4group(p<0.05).Compared with F1group,the content of TNF-a in spinal cord was significantly higher in F4and H4group(p<0.05).Compared with H4group,the content of TNF-α was lower obviously in F4group.Conclusion injection of parecoxib sodium before and after the operation can both cause inhibition on pain;when the dose become bigger the inhibition was more intensive; when giving the same larger doses,the analgesic effect of parecoxib sodium injected before the surgery is better;the analgesia effect may have a relationship with the inhibition of the expression of COX-2and TNF-a in spinal cord.
