Study On Preemptive Analgesia Effecs Of Parecoxib Sodium Of Injection Of IL-6 And COX-2 In The Formalin Test Rats

ObjectivObjective To observe the effects on the blood serum levels of inflammatory factor IL-6In the formalin test Rats and intervation effects of COX-2 specific inhibitor with highcyclooxygenase-2(cox-2)selectivity Parecoxib. explore Parecoxib Sodium the effects andmechanism of preemptive analgesia by determinating serum IL-6 and COX-2 analyse. Toprovide laboratory studies for clinical researching.Methods 65 Waista Rats were randomly divided into four groups,blank group(group kn=5), formalin model group(group F n=20), Preemptive Parecoxib group(group Y n=20),postprocessing Parecoxib group(group H n=20).F group,Y group and H group wassubdiviedInto 4 groups:3 hours group(T3), 6 hours group(T6), 12 hours group(T12),and24 hours group(T24),n=5.Group F was treated with 5% formalin 100ul on iplant .Group Y andGroup H were treated respectively with Parecoxib before and post receiving 5% formalin 100ulon iplant like group F .The blood levels of IL-6 was easured in 3h,6h,12h 24 and after injectionand cox-2 of kidney was easured in 1d,3d,7d 10d. The analysis of variance(ANOVA) of theSSPS software package were used to analyses the data. The differences of these factors betweenthe groups and the correlation between the factors linked to the data were Bonferroni. The valueof P <0.05 was statistically considered .Results In group F, group Y and group H ,IL-6 was significantly higher than that in blankgroup and starting increaing at 3h, peaked at 6h,they have began to descent at 12h,24h wasnormal levels. At 3h,6h, group Y and group H have statistical significance, group H IL-6 wassignificantly higher than that in group Y(P <0. 05). group F and group H have not statisticalsignificance(P >0. 05),At 3h group Y and group K have not statistical significance(P >0.05).(2)COX-2: At 1d,3d,7d group Y and group H have statistical significance(P <0. 05)ï¼›At 1d,10d,group Y and group K have not statistical significance(P >0. 05)ï¼›In group F, groupY and group H , COX-2 was significantly higher than that in blank group and starting increaingat 1d, peaked at 3d,they have began to descent at 7d,10d was normal levels . At 3d, 7d group Yand all group have statistical significance,(P <0. 05)ï¼›But groupY COX-2 was significantlylower than that in group H and was significantly higher than that in group k(P <0. 05);At 1d,3d,group H have all statistical significance(P <0. 05)ï¼›group H COX-2 was significantly lowerthan that in group F; At 7d,group H and group F have not statistical significance(P >0. 05); At10d, group F, group H and group K have statistical significance,(P <0. 05),But group Y groupK have not statistical significance(P >0. 05). ConclusioConclusion In formalin-induced pain model,Preemptive Parecoxib Sodium for injectionhave preemptive analgesia,and the effectIs affirmative;by inhibiting COX-2 to reduceprostaglandin production,thus indirectly inhibiting the increasing of the inflammatory factorIL-6.
