Objective To make emphysema models by active immunization and cigarette smoking usingguinea pigs, Then observe the expression of MMP-9、MMP-12、IP-10 and the destruction ofpulmonary tissue and myocardial tissue in each model,to explore the role of the MMP-9、MMP-12、IP-10 in the emphysema which induced by active immunization.Methods The 24 guinea pigs were randomly divided into three groups, involving activeimmunization group, smoking group and control group. The active immunization group used themethod of subcutaneous injection with compound of elastin fragments and adjuvant at multiplypoints and the smoking group used the traditional method, while the control group performednothing, All thoses animals were bred for 12 weeks. To observe the morphosis of pulmonarytissue and myocardial tissue. Lung tissue slices were stained by hematoxylin-eosin, and thepathologic changes of lung and alveolar radii were determined. and the expression of MMP-9、MMP-12、IP-10 in the pulmonary tissue and myocardial tissue of every model were observed byimmunohistochemical staining. The data were statistically analyzed.Results1.The morphosis of pulmonary tissue and myocardial tissue:The lungs of the smoking groupand the active immunization group showed enlarged lung alveolar airspaces with increasedalveolar radii when compared with the control group(P<0.05, P <0.05).Average lung alveolargap thickness have the obvious difference compared with the control group(P <0.05, P <0.05).2.The expression of MMP-9、MMP-12、IP-10 in the pulmonary tissue and myocardial tissue hasboth significantly increased in the active immunization group and the smoking group , comparedwith the control group (P<0.05).3.The expression of MMP-9、MMP-12 in the pulmonary tissue in the active immunization groupcompared with the smoking group, and there is no obvious difference (P >0.05)。the expressionof IP-10 in the pulmonary tissue has both significantly increased in the active immunizationgroup compared with the smoking group(P <0.05).4. The expression of MMP-9、MMP-12、IP-10 in the pulmonary tissue has both significantlyincreased in the active immunization group compared with the smoking group(P <0.05).Conclusions1. Autoantibodies against lungs were prevalent in guinea pigs with emphysema induced by cigarette exposure. Emphysema model of guinea pig can be established by active immunization.2. MMP-9、MMP-12、IP-10 has important positive influence on emphysema which induced byactive immunization .
