Objective To investigate the influence of antielastin autoimmunity to emphysema,toexplore the expression and significance of Fork head /winged helix protein 3、(Foxp3)、retinoicacid-related orphan receptor)γt(RORγt)and Interleukin-17(IL-17)in Guinea pig model ofemphysema.Methods Smoking and active immunization with subcutaneous injection of elastin peptideswere separately used on guinea pigs emphysema modeling ; Then observe the destruction oflung tissue,and to explore the influence of antielastin autoimmunity to emphysema.Immunohistochemical technique was used to detect the expression of Foxp3、RORγt and IL-17in the pulmonary tissue of guinea pig . The results were compared with the control group by themethod of analysis of variance or kruskal -Wallis test;Spearman rank correlation was used toanalysis the correlation between the Foxp3/RORγt and the expression of IL-17,also used toanalysis the correlation between the ratio of Foxp3/RORγt and the Average radius of alveolar.Result The lung tissue of active immunization group and smoking group has damaged insome extent,and the Average radius of alveolar were significantly longer than the controlgroup, (p<0.05);the ratio of Foxp3/RORγt expression imbalance in smoking group and activeimmunization group , the control group was(1.73±0.72),the smoking group was(0.44±0.09),the active immunization group was(0.51±0.31),expressed as the number ofFoxp3-positive cells decreased, RORγt-positive cells increased,the smoking and activeimmunization group was significantly differently with the control group,(p <0.05); theexpression level of IL-17 in guinea pig,s lung tissue of smoking and active immunization groupwas significantly increased compared with the control group(p <0.05); the difference betweenthe smoking and active immunization group have no significance(p>0.05); The ratio ofFoxp3/RORγt was negatively correlated with the expression level of IL-17 and the Averageradius of alveolar.Conclusion Subcutaneous multi-point injection of elastin peptides can lead to the occurrenceof emphysema in guinea pigs;The ratio of Foxp3 / RORγt in guinea pig’s lung tissue of smokingand active immunization group was imbalanced. This imbalance may lead to abnormaldifferentiation of CD4+T cells,. Autoimmune imbalance may result in chronic obstructivepulmonary disease(COPD), and eventually lead to the deterioration of COPD.
