Objective: The aim of this study is to investigate the expression of My D88 and Rel B in PBMC of patients with acute MS and normal healthy control,in order to explore the correlation and clinical significance between Rel B,My D88 and severity of MS.It provides new ideas and directions for revealing the pathogenesis of MS and seeking new therapeutic targets.Patients and Method:Patients: From June 2018 to January 2019,15 patients with acute MS in the Department of Neurology of the First Hospital of Jilin University were enrolled,including 3 males and 12 females,aged 20-50 years.Gender and age matched HC group included 16 volunteers,including 3 males and 13 females.Method: 1.Inclusion screening: According to the latest MS diagnostic criteria,these patients were diagnosed by neuroimmunological specialists.Meanwhile,16 healthy persons matching with gender and age were selected as control.All patients enrolled in the acute MS group were assessed with the Expanded Disability Status Scale(EDSS),and the relevant clinical data were collected.2.Cell extraction and separation: The 10 ml venous blood was collected from each patient,and the peripheral blood mononuclear cells were separated and extracted by Ficoll method.3.Protein expression: The proteins were extracted from the peripheral blood mononuclear in each group,and the expression of My D88 and Rel B in acute MS group and healthy control group were detected by Western Blot.Furthermore,the software Image Pro-Plus 6.0 was used for quantitative analysis.4.The correlation between the expression of My D88,Rel B and the EDSS score and the annual recurrence rate of(ARR)in patients with acute MS was analyzed.Result: 1.The expression of My D88 in PBMC of patients with acute MS increased significantly compared with healthy control(P=0.000,P<0.05).2.The expression of Rel B in PBMC of patients with acute MS was not significantly higher than that of healthy control group(P=0.694,P>0.05).3.No significant correlation between the expression of My D88 and Rel B in PBMC of acute MS,and EDSS score and ARR could be found.Conclusion: 1.The expression of My D88 in peripheral blood mononuclear cells of patients with acute MS was significantly higher than healthy controls,suggesting that My D88 may be involved in the pathogenesis of MS.2.The expression of Rel B in peripheral blood mononuclear cells of patients with acute MS was not significantly higher than healthy controls,suggesting that Rel B may be not involved in the pathogenesis of MS.3.No significant correlation between the expression of My D88 and Rel B in PBMC of acute MS,and EDSS score and ARR could be found.
