Objective:Research on high glucose environment and join the adiponectin on the myocardial cell proliferation, apoptosis and oxidative stress index ROS effects, To reveal the adiponectin in diabetic cardiomyopathy in protection of myocardial cells.Methods:The cultured myocardial cells divided:normal control group (group N), high glucose group (group G), high glucose+adiponectin group (group A),24h48h were cultured, and72h, using the method of MTT (four methyl thiazolyl blue tetrazolium) cell proliferation was detected by Western blotting assay, cell apoptosis related protein expression of Bax, Bcl-2, DCFH-DA fluorescent probe for detection of myocardial intracellular reactive oxygen species (ROS) level, the cell apoptosis was observed.Results:①MTT method for the detection of human myocardial cell proliferation,24h high glucose group and high glucose+adiponectin group compare difference not to have statistical significance,48h and72h high glucose group compared with controls proliferation decreased significantly (P<0.05), and high glucose+adiponectin group cell proliferation than increased obviously, the difference was statistically significant (P<0.05).②The high glucose group than high glucose group the control group in myocardial cell Bcl-2protein content decreased,Bax protein content increased, adding adiponectin, bcl-2protein content increased significantly, Bax protein content was decreased, the difference was statistically significant (P<0.05).③The expression of ROS in high glucose group than in the normal group, adiponectin expression level of ROS decreased after the intervention, close to the control group, the difference was statistically significant (P<0.05).Conclusions:High glucose sustained effect can inhibit myocardial cell proliferation, increased myocardial cells oxidative stress injury; join adiponectin after intervention, cell proliferation activity increased, reduced on human myocardial cells to oxidative stress injury, can protect human myocardial cells in high glucose environment.
