Objective:Study the effect of adiponectin to proliferation and apoptosis in human renal tubular epithelial cells under high glucose condition and reveal the protective effect of adiponectin in diabetic nephropathy.Methods:After human renal proximal tubular cells were cultured in 5.5mmol/L glucose(normal control group),30.0mmol/L glucose (high glucose group), 30.0mmol/L+2.5μg/ml ADPN (adiponectin group) for 24hs,48hs,72hs separately, MTT assay was also used to detect cell proliferation, and the western blotting detected expression of apoptosis associated Protein Bax, Bcl-2, and DCFH-DA detected ROS level to observe cell apoptosis.Results:①Proliferation in human renal proximal tubular cells in high glucose group were inhibited(P<0.05). It stimulated ROS level and cell apoptosis and were dependent on the time after ADPN(2.5μg/ml) being added(P<0.05).②Proliferation in human renal proximal tubular cells in high glucose group approached normal control. It delayed cell apoptosis and descended ROS level after the postintervention of adiponectin.Conclusions:renal tubular epithelial cells hyperplasia, hypertrophy and apoptosis in high glucose condition are induced with the time prolonged. Adiponectin can promot cell proliferation and delay cell apoptosis.
