Objective:This experiment investigate the traditional Chinese medicine crude extract of Prunella vulgaris and crude extract of Cremastra appendiculata inhibit the proliferation of the thyroid cancer cell in vitro, and observe the50mg/ml of Cremastra appendiculata enhance the proliferative inhibition of SW579with Prunella vulgaris. Besides, we explore the mechanisms of anti-tumor action of Prunella vulgaris and Cremastra appendiculata.Methods:(1)The experiment of proliferative inhibition of SW579:The human thyroid cancer cell line SW579was incubated particularly with different concentrations of crude extract of Prunella vulgaris and crude extract of Cremastra appendiculata. Made the Prunella vulgaris final concentrations were2mg/ml,10mg/ml,20mg/ml,40mg/ml,80mg/ml,200mg/ml and Cremastra appendiculata final concentrations were10mg/ml,50mg/ml,100mg/ml,120mg/ml,150mg/ml,200mg/ml. After incubating24h,48h,72h and96h, the rate of cell proliferation inhibition and IC50were measured by MTT assay. Mixed the50mg/ml remastra appendiculata and the IC50of Prunella vulgaris together and incubated with SW579, then observing both of the two drugs anti-proliferated SW579cell line by MTT assay. It can be divided into four groups:the IC50of Prunella vulgaris,50mg/ml of Cremastra appendiculata, two drugs of IC50of Prunella vulgaris+50mg/ml of Cremastra appendiculata and negative group.(2) Detected the expression of c-myc protein:collected the cell of SW579of each experiment group, The expressions of c-myc protein in different drug therapeutic teams were detected by western blot.Results:(1)In the concentration of10~200mg/ml, the extract of Prunella vulgaris significantly inhibited the growth of SW579cell line, and this effect was dose dependent. The best time was48hours.(2) In the concentration of100~200mg/ml, the extract of Cremastra appendiculata ccould inhibit the growth of SW579cell line, and this effect was dose dependent. The best time was48hours.(3)In the concentration of50mg/ml, Cremastra appendiculata could not inhibit the growth of SW579obviously (P=0.121). So we can put this concentration of Cremastra appendiculata into the IC50of Prunella vulgaris, it could strength the inhibition of SW579(P=0.000)(4)The expression of c-myc protein of the IC50of Prunella vulgaris drug treatment group was lower than the negative group (P=0.000), especially in the tow drugs group (P=0.000). There was no difference between the50mg/ml of Cremastra appendiculata and negative group (p=0.139).Conclusions:(1) In certain range of concentration, the crude extract of Prunella vulgaris and Cremastra appendiculata could anti-proliferate the growth of SW579cell. These effects were dose dependent.(2)50mg/ml of Cremastra appendiculata enhanced the inhibition of IC50of Prunella vulgaris.(3) The expression of c-myc protein was high in thyroid cancer. The anti-proliferation effects of Prunella vulgaris and Cremastra appendiculata on SW579may be connect with inhibition of the c-myc protein.
