Objective: To ivestigate the expression of proline oxidase(POX)in human renalcell carcinoma(RCC)specimens and its relationship with clinicopathologicalcharacteristics.Methods: The SYBR Green I dye based real-time fluorescent quantitative RT-PCR(Real-time RT-PCR)was used to detect the expression of POX mRNA standardand immunohistochemistry(IHC) were performed to evaluate the distribution andexpression of POX protein level in46RCC specimens and adjacent normal tissuecounterparts respectively.Results:The expression of POX mRNA, which was1.27E-02±2.07E-03and3.90E-02±1.01E-02in the RCC specimens and normal tissues respectively, wassignificantly down-regulated in RCC(P<0.01).The positive rates of POX proteinexpression in46RCC specimens(21were expressed at+~+++) was significantlydecreased compared with normal counterparts(36were expressed at+~+++)(P<0.01) from the same patient.. The levels of POX mRNA and protein expressionwere associated with the clinical stage (18cases in stage T1;15cases in stage T2;13cases in stage T3and T4); but not correlated with gender(24male and22femalecases)、age(<50y16cases,≥50y30cases)、tumor size(<7cm29cases,≥7cm17cases)、pathological types(clear cell RCC41cases,chromophobe5cases)andhistological grade(Ⅰ+Ⅱ:26cases,Ⅲ+Ⅳ:20cases)(P>0.05).Conclusions: The POX mRNA and protein expressions are down-regulated inthe human RCC and suggest its possible role in the tumorigenesis of RCC. Theirlower expression are related with the clinical stage,but not concerned with otherclinicopathological features. They may be help to clinical stage and prognosis inRCC.
