Obective To study the expression of phosphorylated stat3(PSTAT3) and vascular endothelial growth fact(VEGF) in renal cell carcinoma and analyze theirroles and relationship between pathological parameters,clinical stage and thedevelopment of renal cell carcinoma(RCC).In order to understand what is the effect and clinical significance of PSTAT3and VEGF during the processof RCC carcinogenesis development and invasion.It maybe provide therapy evidence for gene therapy and antitumor drug research and bring a new effectiveapproach in the treatment of renal neoplasm.Methods Select56specimen of renal cell carcinoma tissues and10specimenof normal renal tissuses.All patients have undergone radical nephrectomy ornephron-sparing partial nephrectomy and without radiotherapy,chemotherapy,immunotherapy and embolization before the surgery.All specimen were made a definite diagnosis by the senior doctor of department of pathology.56cases ofrenal cell carcinoma(36men and20women),According to2002AJCC(American Joint Committee on Cancer Staging) TNM staging:stage T121cases,stage T215cases,stage T314cases,stage T46cases.According to1997Fuhrmangrading:grade I~Ⅱas well differentiated25cases,grade III as moderately differentiated20cases and grade Ⅳ as poorly differentiated11cases. the expression of PSTAT3and VEGF were detected by the immunohistochemical staining and investigating the relationship between PSTAT3and VEGF.experimentaldata is analysised by SPSS13.0software.P<0.05to have significant differences as to inspection standards.Results The positive rate of PSTAT3and VEGF in RCC tissues is67.86%and64.29%,while in the normal kidney tissues is10%and20%.The positive rate of PSTAT3and VEGF in RCC tissues were both higher than those in no rmal renal tissues and the differences were significant statistically (p<0.05);The expression of PSTAT3and VEGF in poorly differentiated RCC tissues weresignificantly higher than those in well differentiated RCC Tissues(p<0.05).PSTAT3and VEGF proteins in TNM stageⅢ+Ⅳ were significantly higher than those of TNM stage Ⅰ+Ⅱ and PSTAT3and VEGF proteins in RCC withlymph node metastases also higher than those in RCC without lymph node metastases(P<0.05).there was no statistics significance of the expression of PSTAT3and VEGF between the different age and gender group.Spearman correlation analysis shown that PSTAT3was positively related to VEGF in renal carcinoma(r=0.770,P<0.001).Conclusion Increased expression of PSTAT3and VEGF correlate with the carcinogenesis and progression of RCC.The high expression of PSTAT3and VEGF correlate with the angiogenesis,invasion and metastasis of RCC.The high expression of PSTAT3and VEGF correlate with the clinical stage,lymph nodesmetastasis and cell differentiation of RCC,while there is no correlation withage and gender.PSTAT3was positively related to VEGF in renal carcinoma(r=0.770,P<0.01).STAT3may regulate the expression of VEGF,So it could bea potential target for inhibiting VEGF expression and angiogenesis for the treatment of RCC.They could be treated as a predictable fact of the prognosisof renal carcinoma.
