Objective Using small interfering RNA(siRNA)to decrease the expressing of Kiss1(kiss1)gene in 786-O and ACHN renal cells lines.And to investigate the role of Kiss1 siRNA(si-Kiss1)on renal cell angiogenesis.Methods In vitro: First,Immunohistochemistry was used to assess the expression of Kiss1 in 342 renal cancer tissues and 46 normal renal tissues,and its correlation with clinicopathological features and prognosis of renal cancer were also analyzed;Using Kiss1 small interfering RNA(siRNA)which was synthesized chemically in vitro to transfect 786-O and ACHN renal cancer cells and to investigate the effects of Kiss1 siRNA on Kiss1 and VEGF expression in 786-O and ACHN cells by Western Blot.48 hours later,we observe that how many tubes formate through tube formation assay?Results In vitro,(1)IHC showed that the expression of Kiss1 was decreased significantly in RCC tissue compared with NRT(P<0.05)and it was correlated with renal carcinoma pT status(P<0.05),Kiss1 expression has no correlation with the survival of patients.(2)Western Blot showed that the Kiss1 expression is decreased after Kiss1 knockdown in both 786-O and ACHN renal carcinoma cells.(786-O and ACHN decreased77.7% and 68.1%,P<0.001,n=3);Western Blot showed that the VEGF expression is increased after Kiss1 knockdown in both 786-O and ACHN renal carcinoma cells.(786-O and ACHN increased56.2% and 62.1%,P<0.001,n=3)(3)Conclusion We founded that Kiss1 staining was decreased significantly in RCC tissue compared with NRT.Knockdown of Kiss1 leads to inhibition of Kiss1 and increase the ability of VEGF expression and angiogenesis.
