Objective: To compare the effects of perilla oil and fenofibrate on blood lipid andvessels in hyperlipidemia.Methods:(Ⅰ) Basic Research: Forty(8weeks-old,male)Sprague Dawley rats wererandomly and evenly divided into five groups:normal control(normal diet),modelcontro(lhigh-fat diet),fenofibrate group(high-fat diet+100mg/Kg d fenofibrate),perillaoil low-dose treatment group(high-fat diet+0.34g/Kg d perilla oil),perilla oil high dosetreatment group(high-fat diet+2.00g/Kg d perilla oil)。After12weeks of feeding,therats were sacrificed for detection of the levels of lipid、interleukin-6(IL-6)。Wall tolumen area ratio (W/L) of mesenteric artery(3rd grade branch) were assessed afterhematoxylineosin staining。Vascular reactivity to vasoactive substance were recordedusing mesenteric artery rings,which was solated from rats by automatic balancerecording meter。(Ⅱ) Clinical Research:60cases of patients with hyperlipidemia wererandomly divided into two groups,treating with perilla oil soft capsule and fenofibraterespectively for8weeks。Blood lipids ingredients、liver function、brachial-anklepulse wave velocity(baPWV)、ankle brachial index(ABI)、large artery elasticity(C1)、small artery elasticity(C2)were measured in the two groups before and aftertreatment。Results:(Ⅰ) Basic Research:(1)After feeding12weeks,the rat weight of modelcontrol and perilla oil low-dose treatment group were significantly higher than that innormal control (P<0.05),when fenofibrate group and perilla oil high dose treatmentgroup were lower than model control(P<0.05)。(2)Total Cholesterol(TC)、lowdensity lipoprotein cholesterin(LDL-C)、triglycercide(TG)、very lowdensitylipoprotein cholesterol(VLDL-C) and IL-6in model control were significantly higher than normal contro(lP<0.05);perilla oil low and high-dose treatment all couldreduce Blood Lipid and IL-6,and the effects on LDL-C、TG、VLDL-C exerted thedose-dependent manner(P<0.05)。No significantly difference was founded betweenperilla oil high dose treatment group and fenofibrate group(P>0.05)。(3)In modelcontrol,the W/L was much more higher and the relaxation sensitivity to sodiumnitroprusside was significantly decreased than normal control(P<0.01),perilla oilcould improved on W/L (P<0.05) and relaxation sensitivity to sodiumnitroprusside(P<0.01)by dose-dependent,without significantly difference to normalcontro(lP>0.05)。(Ⅱ)Clinical Research:(1)Triglyceride was significantly decreasedin both of the two groups,and the fenofibrate group was much more remarkable(30.4%VS20.7%,P<0.05)。(2)baPWV、C1and C2were dropped dramatically,without Significantly Differences between perilla oil soft capsule group andfenofibrate group。(3)Two cases(6.7%)of patients showed up mild hepaticdysfunction in fenofibrate group,and recoverded normal after altered to perilla oil softcapsule, d none of patients treated with perilla oil soft capsule had hypohepatia。Conclusion: Perilla oil may ameliorated dyslipidemia,reduced inflammatoryfactor,attenuated vascular hypertrophy,improved the vessels function of the hyperlipidemia,and the curative effets were same to fenofibrate,without hypohepatia。...
