Objective:To investigate the possible association and to reveal the roles betweenApolipoprotein A1(ApoA1), Apolipoprotein C3(ApoC3) gene polymorphism andHypertensive disorder complicating pregnancy(HDCP),and to provide the scientificbasis for the prevention and treatment of the clinical patients with HDCP.Methods:Examined the distribution of4known SNPs(-75G>A,+83C>T in ApoA1;-455T>C,-482C>T in ApoC3) between247patients with HDCP(the case group) and295normal controls(the control group) by PCR-RFLP. Then analysed the associationof these SNPs with plasma lipid levels of HDCP.Results:1.There were no significant difference in age, height, weight, and gestational age betweenthe case group and the control group (P>0.05);the frequencies of different genotypesof the4known SNPs were in agreement with Hardy-Weinberg equilibrium in ourstudy(P<0.05).2.The genotypes and allele frequencies with4SNPs in the control group:The frequencies of ApoA1-75G>A GG,GA,AA genotypes were49.8%,38.6%,11.5%, allele frequencies were G69.2%,A30.8%; the frequencies oftheApoA1+83C>T CC,CT,TT genotypes were89.2%,10.5%,0.3%, allele frequencieswere C94.4%,T5.6%;the frequencies of the ApoC3-482C>T CC,CT,TT genotypes were29.8%,51.9%,18.3%, allele frequencies were C55.8%,T44.2%; thefrequencies of the ApoC3-455T>C CC,CT,TT genotypes were15.6%,54.6%,29.8%,allele frequencies were C42.9%,T57.1%.3.The genotypes and allele frequencies with4SNPs in the case group:The frequencies of ApoA1-75G>A GG,GA,AA genotypes were44.1%,44.1%,11.7%, allele frequencies were G66.2%,A33.8%; the frequencies of theApoA1+83C>TCC,CT genotypes were90.3%,9.7%, allele frequencies were C95.1%,T4.9%;thefrequencies of the ApoC3-482C>T CC,CT,TT genotypes were32%,51.4%,16.6%,allele frequencies were C57.7%,T42.3%; the frequencies of the ApoC3-455T>CCC,CT,TT genotypes were17.6%,51.0%,31.4%, allele frequencies were C43.1%,T56.9%.4.There were no significant difference in genotypes and allele frequencies with4SNPs in the two groups statistic analysised by SPSS17.0(P>0.05).5. The association between the4SNPs and the plasma lipid levels of HDCP:There were significant difference in plasma ApoA1level in three genotypes ofApoA1-75G>A, and the plasma ApoA1level with G/A genotype in case group wasthe highest.(X2=6.409,P=0.041). There were significant difference in plasmaApoA1level between the C/T and the T/T genotype of ApoC3-482C>T (P=0.033).There were significant difference in plasma TG level in three genotypes ofApoC3-455T>C, and the plasma TG level with C/C genotype in case group was thehighest.(X2=9.068,P=0.011).6.Haplotype analyses with Shesis sofeware demonstrated that2SNP haplotype-482T≡-455T was a weak protective factor for HDCP, OR0.452.7. There were significant difference in the TG,ApoA1,ApoB, HDL-c levels andMBI between the case group and the control group.Conclusion:The study found that four kinds of single nucleotide polymorphisms ofApoliprotein A1,C3gene were existed in the Han Chinese women in Fujian Province.Hyperlipidemia and obesity were exsited in patients with HDCP. There might be anassociation between polymorphism in the Apoliprotein A1,C3gene with plasma lipidlevels of Hypertensive disorder complicating pregnancy, and affected the plasma lipid levels.2SNP haplotype-482T≡-455T was a weak protective factor for HDCP. Theexact mechanism of such relationship was still unclear, and was needed to study.
