Objective Quantitative analysis of Epstein-Barr virus(EBV) DNA levels in saliva,blood serum and peripheral blood cells(PBCs) of nasopharyngeal carcinoma(NPC)patients,to investigate the relationship between EBV-DNA levels and NPC.Methods EBV-DNA in Saliva,Blood serum and PBCs of40patients withprimary NPC and50controls were quantitative detected by using real-timequantitative PCR (FQ—PCR).Results The detected rate of blood serum EBV-DNA in patients with NPC (67.5%)was significantly higher than that in controls(2%)(2=44.48,P<0.001). TheEBV-DNA detected rate in Saliva was no significant difference with that in PBCs,both patients with NPC and controls. The EBV-DNA level in Saliva was correlatedto the EBV-DNA load in PBCs,both patients with NPC and controls.In patients withNPC,the EBV-DNA level in blood serum was significantly increased along with theTNM stage.Conclusion Epstein-Barr virus long-standing in most people’s saliva,may alsoexist in the circumference blood B lymphocyte,it is not a kind of precancerousstate.The nasopharyngeal epidermis which has heterogeneously changed and beinginfected EB virus can be regarded as the precancerous change.The EBV-DNAexamination of blood serum has the high sensitivity (67.5%) and the high specificity(98%) in nasopharyngeal cancer’s clinical diagnosis,may be used in pharyngealcancer’s early diagnosis. The EBV-DNA of Blood serum possibly originate fromtumor DNA and will improve TNM staging in NPC’s clinical practice on molecularlevel.
