Objective:To detect the expression of PD-1 in peripheral blood T cells and to explore the relationship between PD-1 and clinical features and short-term efficacy of patients with nasopharyngeal carcinoma.METHODS:Fifteen patients with nasopharyngeal carcinoma who were admitted to the Tumor Hospital of Xinjiang Medical University from January 2015 to April 2016 and 10 healthy controls were enrolled.Flow cytometry was used to detect 10 healthy controls in peripheral blood T cells expression level and expression of PD-1 in peripheral blood T cells at different time points before and after treatment in 15 patients with nasopharyngeal carcinoma.Comparison of PD-1expression levels in peripheral blood T cells between healthy controls and nasopharyngeal carcinoma patients.Analysis of nasopharyngeal carcinoma The correlation between the expression of PD-1 in peripheral blood T cells and clinical features was observed.The dynamic changes of PD-1 expression in peripheral blood T cells before and after treatment of nasopharyngeal carcinoma were observed.Results: 1.The percentage of CD4+ T cells in peripheral blood of nasopharyngeal carcinoma was(46.49 ± 13.263)%,which was significantly lower than that of healthy controls(54.08±14.024)%,but the difference was not statistically significant.The percentage of CD8+ T cells in peripheral blood of nasopharyngeal carcinoma was(43.11± 14.024)%,which was significantly higher than that of healthy controls(33.76 ± 6.841)%.The difference was statistically significant(P<0.05).The expression rate of PD-1 in peripheral blood CD4+ T cells of nasopharyngeal carcinoma(17.71±4.576)% was significantly higher than that of healthy controls(13.02±4.286)%(P=0.017),and the expression rate of PD-1 in CD8+ T cells.(18.48±7.183)% was also significantly higher than the healthy control group(12.22±1.614)%(P=0.005),and the difference was statistically significant.3.The expression rate of PD-1 in peripheral blood CD4+ T cells is related to smoking history.The expression rate of PD-1 in CD4+ T cells in the smoking group was higher than that in thenon-smoking group.There was no significant correlation between the expression of PD-1in CD4+T and CD8+ T cells and clinical factors such as age,PLR,NLR,BMI,HB,LDH and EBV DNA concentration.There was no significant correlation between the expression rate of PD-1 in CD4+ T cells(rs=0.248,P=0.372)and CD8+ T cells(rs=-0.108,P=0.701)and the short-term efficacy of locally advanced nasopharyngeal carcinoma.With the passage of time,the expression rate of PD-1 in nasopharyngeal carcinoma CD4+ T cells increased first and then decreased with time,and the difference was statistically significant(P<0.05).Conclusion: The expression rate of PD-1 in peripheral blood of patients with nasopharyngeal carcinoma has a certain correlation with its clinical features,but the relationship between efficacy and prognosis needs to be further determined.
