Objective: The study was to investigate the effect of adiponectin on the expression of kisspeptin and regulation of KISS1gene transcription in the ARC of SD rats. We also investigated whether adiponectin regulated the expression of kisspeptin through signal pathway of AMPK.Methods:1. There were40ovariectomized SD rats with oral administration of NILE. We randomly assigned them into four groups, which were respectively given intracerebroventricular injection of adiponectin, AMPK excitomotor AICAR, AMPK inhibitor Compound C and contraposition.2. ARC in the hypothalamus of female rate were observed and analyzed by Immunohistochemistry stain, as well as the number of kisspeptin‐ir cells was accounted.3. The difference of Kiss1mRNA among four groups was detected by Realtime PCR.Results:1. Inhibition of AMPK by Compound C, a relatively specific AMPK inhibitor, augmented KISS1mRNA expression and the kiss1‐ir cells at ARC in the hypothalamus of female rats.2. There was also a trend that adiponectin, AICAR (AMPK excitomotor)could inhibit the numbers of the kiss1‐ir cells and the expression of KISS1mRNA.Conclusion:Adiponectin could inhibit the expression of kiss1gene at ARC in the hypothalamus of female rats through AMPK pathway.
