Objective: In this study,the pathological changes in the hypothalamus and the diff erences in the protein expressions of Kisspeptin,GPR54,and GnRH in the hypotha-lamus were explored to explore the effects of overweight on the reproductive function of rats and the reversal effect of Zuogui Pill.Methods: Eighty female rats with regular estrous cycles and suitable for running training were selected for experimental study.Randomly divide them into A and B groups,and set up the blank group(A1,B1),the model group(A2,B2),the diet recovery group(A3,B3),and the diet recovery+zuogui pill group(A4,B4)are set up.A total of 8 small group of 10 rats.Rats in the A2-A4 groups were modeled by "L-carnitine+half diet+excessive exercise" for 4 weeks,and the rats in the A1 and A2 groups were sacrificed immediately after the modeling,and the rats in the A3 and A4 groups returned to the normal diet.The A4 group was treated with Zuogui Pill at the same time,and the rats in the A3 and A4 group were sacrificed4 weeks later to collect indicators.The modeling time of group B was 8 weeks,and the experimental method was the same as that of group A.Result: 1.Weight change: rats lost 4% of body weight at 4 weeks after modeling and 23% at 8 weeks of modeling.The difference between the two groups was statistically significant(P<0.01).2.Changes in estrous cycle: the estrous cycle disturbance rate in the model group was 100%;The estrous cycle disturbance rates in the A3 and A4 groups were 62.5% and 44.4%,respectively,which were significantly lower than those in the A2 group(P<0.05);The rates of estrous cycle disturbances in the B3 and B4 groups were 88.9% and 77.8%,respectively,respectively.Compared with the B2 group,the differences were not statistically significant(P>0.05).3.Changes in serum sex hormone levels: compared with the blank group,the serum FSH and LH values of the model group increased,and the E2 value decreased.The differences between the two groups were significant(P<0.01);A3,A4 and A2 or B3,B4 and B2 compared,FSH,LH decreased and E2 increased,the difference was statistically significant(P<0.01);compared with A4 and A3 groups,FSH andLH decreased significantly(P<0.01),and E2 levels increased significantly(P<0.05);Compared with B3 and B4 groups,the difference was not statistically significant(P>0.05).Compared with the A2 group,the FSH and LH of the rats in the B2 group increased and the E2 decreased more significantly(P<0.05).4.Effect on the morphology of hypothalamus:in the model group,hypothalamus neurons have varying degrees of degeneration or necrosis,the number decreases,the volume becomes smaller,the nucleus shrinks or dissolves,and the surrounding space widens;The B2 group had more severe lesions in the hypothalamus than the A2 group,with microglia proliferation;After normal diet and Zuogui pill intervention,the tissue lesions improved,and the glial cell proliferation was seen in a larger volume.The improvement of the diet + Zuogui Wan group was relatively obvious.5.Effect on the expression of Kisspeptin,GPR54,and GnRH proteins in the hypothalamus: the expression levels of Kisspeptin,GPR54,GnRH protein in the model group was significantly reduced compared with the blank group(P<0.01),and the reduction was more significant in the B2 group,and the difference was significantly different from the A2 group(P<0.01);The expression levels of Kisspeptin,GPR54,and GnRH in A3 and A4 groups were increased compared with A2 group rats(P<0.01),and A4 increased more significantly than that in A3 group(P<0.01);The expressions of Kisspeptin,GPR54,and GnRH in B3 and B4 were increased compared with B2 rats,but the difference was not statistically significant(P>0.05),and the difference between B4 and B3 groups was not statistically significant(P>0.05).Conclusions: 1.Serum FSH and LH are increased,E2 is decreased,and the estrous cycle is disordered,which indicates that the rats’ reproductive dysfunction isabnormal;2.Excessive weight loss caused by malnutrition causes damage to the rat’shypothalamus;3.Excessive slimming rats hypothalamic Kisspeptin,GPR54,GnRH expression levels decreased,suggesting that the Kisspeptin/GPR54 system may be related to excessive weight loss caused by reproductive dysfunction in rats;4.Zuo Gui Pill may regulate the secretion of sex hormones by promoting the Kisspeptin/GPR54 system and promote hypothalamic tissue lesions effect of repairing and improving reproductive function in rats.
